荆防颗粒治疗2,4-二硝基氯苯诱导小鼠迟发性过敏反应药效评价研究

摘要 目的 探讨荆防颗粒对2,4-二硝基氯苯(2,4-dinitrochlorobenzene,DNCB)诱导的迟发性过敏反应(delayed type hypersensitivity,DTH)小鼠模型的药效评价研究。方法 分别设立对照组,模型组,醋酸泼尼松组(5 mg·kg-1),荆防颗粒高、中、低剂量(23.2、11.6、5.8 g·kg^(-1))组,各组均用DNCB诱导(对照组除外)。在DNCB诱导后当天开始灌胃给药,观察体质量、耳廓肿胀度、背部皮损、脾指数变化;HE染色和甲苯胺蓝染色分别观察耳组织病理形态学改变和背部皮损组织肥大细胞脱颗粒;选择时间点记录模型小鼠搔抓次数;酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测血清中免疫球蛋白E (immunoglobulin E,IgE)、白细胞介素-4 (interleukin-4,IL-4)、白细胞介素-9(interleukin-9,IL-9)、白细胞介素-17(interleukin-17,IL-17)以及干扰素-γ(interferon-γ,IFN-γ)水平的变化。结果 与对照组比较,模型组搔抓次数增多且背部出现皮肤水肿和红斑、结痂,右耳肿胀度增加,脾脏指数明显升高(P<0.01),耳组织HE染色可见淋巴细胞、中性粒细胞浸润,血清中IgE、IFN-γ、IL-4、IL-9、IL-17表达明显升高(P<0.01);与模型组比较,醋酸泼尼松组和荆防颗粒各剂量组皮肤水肿和红斑、结痂与搔抓均有减轻,耳廓肿胀度减轻,炎症细胞浸润明显减少,脾脏指数以及血清中IgE炎症因子水平降低,以醋酸泼尼松组和荆防颗粒高剂量组最为明显(P<0.01)。结论 荆防颗粒能够通过降低相关炎症细胞因子同时增强模型小鼠的免疫功能对迟发性过敏反应小鼠发挥治疗作用,是潜在治疗迟发性过敏反应的药物。 Objective To evaluate the efficacy of Jingfang Granule(荆防颗粒)on 2,4-dinitrochlorobenzene(DNCB)-induced delayed type hypersensitivity(DTH)in mice.Methods Control group,model group,prednisone acetate group(5 mg·kg^(-1)),Jingfang Granule high,medium and low dose groups(23.2,11.6,5.8 g·kg^(-1))were set up respectively.All groups were induced with DNCB except the control group.On the day after DNCB stimulation,the corresponding drugs were given to each group.After treatment,the changes of back skin lesions,body weight,auricle swelling and spleen index were observed.HE staining and toluidine blue staining were used to observe the pathological changes of ear tissue and degranulation of mast cells in back skin lesions.The scratching times of model mice were recorded at selected time points.The levels of IgE,IL-4,IL-9,IL-17 and IFN-γin serum were detected by ELISA.Results Compared with the control group,the skin lesions of the model group showed edema,erythema and scab with scratching,the swelling degree of the right ear increased,and the spleen index increased significantly(P<0.01).The infiltration of lymphocytes and neutrophils in the ear tissue was observed by HE staining.The expressions of IgE,IFN-γ,IL-4,IL-9 and IL-17 in serum were significantly increased(P<0.01).Compared with the model group,the erythema,scale and scab of skin lesions on the back of prednisone acetate group and Jingfang Granule groups were alleviated to different degrees,the degree of auricle swelling was alleviated,the infiltration of inflammatory cells was significantly reduced,the spleen index and the content of IgE inflammatory factors in serum were decreased.The prednisone acetate group and Jingfang Granule high dose group were the most obvious(P<0.01).Conclusion Jingfang Granule can play a therapeutic role by inhibiting the expression of related inflammatory factors and regulating the immune function of mice with DTH,which is a potential drug for the treatment of DTH.
