组织透明化技术在神经生物学领域的发展和应用

摘要 组织透明化技术自问世以来,不仅为脑科学领域在研究全脑结构、神经系统、神经环路、特征蛋白等方面提供了一种更加有效的工具,而且已经实现了对大脑亚细胞分辨率成像以及远距离追踪单个神经元和轴突。这些对于破译健康大脑的功能结构和神经病理机制具有重要意义。在脑组织三维成像方面,传统的组织成像技术由于成像深度和透光度的限制无法呈现完整的三维立体结构,而组织透明化技术能够通过组织清除试剂实现大脑透明化,并与成像技术及神经示踪技术相结合,提供完整无损的组织3D图像。未来深入发展组织透明化技术并结合其他脑科学技术,可拓展其应用广度和研究深度。 Since its advent,tissue transparency technology has not only provided a more effective tool for brain science to study the whole brain structure,nervous system,neural circuits,characteristic proteins,etc.,but also achieved subcellular resolution imaging of the brain and remote tracking of individual neurons and axons.These are important for deciphering the functional structure and neuropathological mechanism of the healthy brain.In three-dimensional imaging of brain tissue,the traditional tissue imaging technology cannot present a complete three-dimensional structure due to the limitations of imaging depth and light transmittance,while tissue transparency technology can achieve brain transparency through tissue clearance reagents,and in combination with imaging and nerve tracer technique,provide intact 3D tissue images.In the future,further development of tissue transparency technology combined with other brain science technologies can expand its application width and research depth.
