摘要
本研究以西番莲叶为原料,采用热水提取和碱法提取得到水提和碱提西番莲叶粗多糖,采用DEAE-52纤维素柱层析分离,得到WPEL-A和APEL-A两种多糖,并进行化学组成分析、结构表征和体外抗氧化活性研究。结果表明,分离得到的WPEL-A和APEL-A的总糖含量分别为70.31%、79.23%,WPEL-A的分子量分布为3.65×10^(4)和2.26×10^(3)Da,APEL-A的分子量分布为8.53×10^(5)、1.13×10^(5)和1.66×10^(3)Da。WPEL-A主要由半乳糖、半乳糖醛酸和阿拉伯糖组成,APEL-A主要由葡萄糖、阿拉伯糖和半乳糖组成。扫描电镜结果表明,WPEL-A相对APEL-A破碎更明显,颗粒更小且较均一。体外抗氧化结果表明,两种多糖都具有一定自由基清除能力和还原能力。由于WPEL-A分子量较小且半乳糖醛酸含量较高,而具有更强的抗氧化活性,多糖浓度为3 mg/mL时,DPPH自由基清除率、还原能力、羟基自由基清除率和超氧阴离子自由基清除率分别为70.61%、0.75、21.90%和18.55%。本研究可为西番莲叶多糖的开发利用和生物活性的深入研究提供实验基础。
Passiflora edulis Sims leaf was used as raw material,and the crude polysaccharides from water-extracted and alkali-extracted Passiflora edulis Sims leaf were obtained by hot water extraction and alkali extraction.Two polysaccharides,WPEL-A and APEL-A,were obtained by DEAE-52 cellulose column chromatography and the chemical composition analysis,structural characterization and in vitro antioxidant activity studies were conducted.The results showed that the total sugar content of WPEL-A and APEL-A were 70.31% and 79.23%,respectively.The molecular weight distributions of WPEL-A were 3.65×10^(4) and 2.26×10^(3) Da,respectively,and those of APEL-A were 8.53×10^(5) Da,1.13×10^(5) Da and 1.66×10^(3) Da.WPEL-A was mainly composed of galactose,galacturonic acid and arabinose,APEL-A was mainly composed of glucose,arabinose and galactose.The results of scanning electron microscopy showed that the fragmentation of WPEL-A was more obvious than that of APEL-A,and the particles were smaller and more homogeneous.The in vitro antioxidant results showed that both polysaccharides had certain free radical scavenging and reducing abilities.WPEL-A has stronger antioxidant activity due to its smaller molecular weight and higher galacturonic acid content.When the polysaccharide concentration was 3 mg/mL,the DPPH free radical scavenging rate,reducing ability,hydroxyl radical scavenging rate and superoxide anion free radical scavenging rate were 70.61%,0.75,21.90%and 18.55%,respectively.This study can provide an experimental basis for the development and exploitation of the polysaccharides from Passiflora edulis Sims leaves and the in-depth study of their biological activities.
