哈巴苷通过Wnt/β-catenin通路改善大鼠脑缺血再灌注损伤

Harpagide improves cerebral ischemia-reperfusion injury in rats through Wnt/β-catenin pathway

摘要 目的探究哈巴苷干预大鼠脑缺血再灌注损伤的作用及其对Wnt/β-catenin信号通路的影响。方法18只雄性SD大鼠随机分为假手术组、模型组和哈巴苷干预组,每组6只,对各组大鼠进行神经功能评估。再灌注7d后,TTC染色法检测大鼠脑梗死体积;干湿重法检测大鼠脑组织含水量;TUNEL染色检测大鼠脑组织神经细胞凋亡;荧光探针检测大鼠脑组织神经细胞ROS水平;Western blot检测大鼠脑组织β-catenin和activeβ-catenin表达水平。结果与对照组相比,模型组大鼠神经功能评分下降,脑梗死体积和脑含水量增加。加入哈巴苷干预后,大鼠神经功能评分提高,脑梗死体积减小,脑水肿减轻。与对照组相比,模型组大鼠神经细胞凋亡率增加,活性氧水平增加;加入哈巴苷干预后,大鼠神经细胞凋亡率下降,活性氧释放减少。与对照组相比,模型组大鼠β-catenin和activeβ-catenin表达水平提高,加入哈巴苷后其表达水平进一步升高,Wnt/β-catenin信号通路被激活。结论哈巴苷能够通过激活Wnt/β-catenin信号通路减轻缺血再灌注导致的大鼠脑损伤。 Objective To investigate the effect of harpagide on cerebral ischemia-reperfusion injury in rats and its effect on Wnt/β-catenin signaling pathway.Methods Eighteen male SD rats were randomly divided into sham operation group,model group and harpagide intervention group,with 6 rats in each group.Neurological function was evaluated in each group.After 7 days of reperfusion,the volume of cerebral infarction was detected by TTC staining.The water content in brain tissue of rat was detected by dry and wet weight method.TUNEL staining was used to detect the apoptosis of nerve cells in brain of rats.Fluorescence probe was used to detect level of ROS in brain of rat.The expressions ofβ-catenin and activeβ-catenin in rat brain were detected by Western blot.Results Compared with the control group,the neurological function score of rats was decreased,and the volume of cerebral infarction and the cerebral water content were increased in the model group.After the addition of harpagide,the neurological function score of rats was promoted,the volume of cerebral infarction and the cerebral water content was reduced.Compared with the control group,the neuronal apoptosis rate and death rate of nerve cells in the model group were increased,and the level of ROS was promoted.After the addition of harpagide,the apoptosis and necrosis of nerve cells in rats was decreased,and the release of ROS was decreased.Compared with the control group,the expression ofβ-catenin and activeβ-catenin in the model group were increased,and the expression level ofβ-catenin and activeβ-catenin was further increased in Harpagide group,indicating that Wnt/β-catenin signaling pathway was activated under harpagide treatment.Conclusion Harpagide can reduce brain injury induced by ischemia-reperfusion in rats by activating Wnt/β-catenin signaling pathway.
