摘要
目的探讨电针结合经颅脉冲电刺激促进脑卒中大鼠血管新生的作用机制。方法60只SD大鼠被随机分为假手术组(sham)、模型组(model)、电针组(EA)、经颅脉冲电刺激组(tPCS)、电针结合经颅脉冲电刺激组(EA+tPCS)及EA+tPCS+miRNA-150 inhibitor组(miRNA-150 inhibitor),每组10只。采用线栓法制备慢性脑卒中模型。对各组大鼠神经行为学进行评分;HE染色观察大鼠脑皮质区病理损伤;免疫荧光染色测定大鼠脑组织微血管密度;qRT-PCR检测脑组织中血管内皮生长因子(VEGF)、血管生成素-2(Ang-2)mRNA及miRNA-150表达水平;Western blot检测VEGF蛋白表达。结果与模型组相比,EA、tPCS及EA+tPCS组大鼠神经行为学评分显著降低,大鼠脑皮质病理损伤明显减轻,脑组织微血管密度显著增加,VEGF和Ang-2 mRNA表达水平显著升高,miRNA-150表达水平显著升高,且VEGF蛋白表达水平亦显著增加,上述结果尤以EA+tPCS组的效果最为显著。然而,当大鼠注射miRNA-150 inhibitor后,EA+tPCS的改善作用均被削弱或抑制。结论EA联合tPCS改善脑卒中大鼠神经功能的作用机制可能与上调miRNA-150进而调控VEGF表达,促进血管新生有关。
Objective To investigate the mechanism of electroacupuncture combined with transcranial pulsed current stimulation to promote angiogenesis in stroke rats.Methods 60 SD rats were randomly divided into sham group,model group,electroacupuncture group(EA),transcranial pulsed current stimulation(tPCS),EA combined with tPCS group(EA+tPCS),and EA+tPCS+miRNA-150 inhibitor group(miRNA-150 inhibitor)with 10 rats in each group.The model of chronic stroke was prepared by thread occlusion method.Neurobehavioral scores of rats in each group were detected.The pathological damage of cerebral cortex were observed by HE staining.The microvascular density was measured by immunofluorescent staining.The expressions of vascular endothelial growth factor(VEGF),angiopoietin-2(Ang-2)mRNA and miRNA-150 were detected by qRT-PCR,and the expression of VEGF was detected by Western blot.Results Compared with model group,neurobehavioral scores of rats in EA,tPCS and EA+tPCS groups were significantly decreased,pathological damage of cerebral cortex was significantly reduced,microvascular density of brain tissue was significantly increased,the levels of VEGF and Ang-2 mRNA were significantly increased,and the level of miRNA-150 was significantly increased.The expression of VEGF was also significantly increased,especially in the EA+tPCS group.However,when the rats were injected with miRNA-150 inhibitor,the improvement effect of EA+tPCS was weakened or inhibited.Conclusion The mechanism of EA combined with tPCS to improve nerve function in stroke rats may be related to the up-regulation of miRNA-150,thus regulating the expression of VEGF and promoting angiogenesis.
