摘要
目的探讨二甲双胍与电针对创伤性脑损伤(TBI)的治疗作用。方法以C57BL/6小鼠为研究对象,分为对照组(Control),TBI组,TBI+E(Electroacupuncture,E)组,TBI+M(Metformin,M)组和TBI+E+M组。对照组仅摘除颅骨,TBI组实施重物坠落法造成脑损伤,TBI+E组在损伤后第二天以电针治疗,TBI+M组在损伤后第二天以二甲双胍治疗,TBI+E+M组在损伤后第二天以电针和二甲双胍联合治疗。在TBI后7d,通过新物体识别试验检测认知功能,通过组织病理学实验检测组织缺损、小胶质细胞的募集活化和髓鞘再生。结果TBI+E+M组小鼠认知功能恢复明显强于TBI组,TBI组出现组织缺损、皮层空洞;TBI+E组、TBI+M组和TBI+E+M组小胶质细胞数目均出现降低,TBI+E+M组恢复到对照组水平,小胶质细胞的募集活化受到明显抑制;TBI+E+M组髓鞘含量明显多于TBI+E组和TBI+M组,髓鞘再生能力明显增强。结论二甲双胍联合电针治疗脑损伤改善小鼠认知功能障碍,抑制小胶质细胞的募集活化和促进髓鞘再生。
Objective To investigate the effect and significance of metformin and electroacupuncture on the treatment of traumatic brain injury.Methods C57BL/6 mice were divided into control group(only the skull was removed),TBI group,TBI+E(Electroacupuncture,E)group,TBI+M(Metformin,M)group and TBI+E+M group.Seven days post⁃TBI,cognitive function was detected by new object recognition test,and tissue defects,microglia recruitment activation and remyelination were examined by HE.Results The cognitive function recovery was significantly better in TBI+E+M mice than in TBI+E group and TBI+M group.The number of microglia was higher in TBI+E+M group than in TBI+E and TBI+M group.The myelin content was significantly increased in TBI+E+M group than in TBI+E group and TBI+M group.Conclusion Metformin+electroacupuncture improved cognitive dysfunction,which is related to inhibiting the activation of inflammatory cell microglia and promoting remyelination in C57BL/6 TBI mice.
