摘要
目的探讨TANK结合激酶1(TBK1)在高级别脑胶质瘤髓样细胞中的表达特征.方法选取2019年4月至2021年2月就诊于河北医科大学第二医院的27例WHO分级标准为高级别胶质瘤患者作为研究对象,采用磁珠分选的方法检测胶质瘤瘤旁和肿瘤组织中整合素αM(CD11b)阳性髓样细胞中TBK1的表达,免疫荧光染色观察肿瘤微环境巨噬细胞中磷酸化TBK1(p-TBK1)的表达,并通过流式细胞分析患者术前1 d、术后10~14 d外周血经典单核细胞中p-TBK1的表达变化.结果高级别胶质瘤肿瘤组织CD11b阳性细胞中TBK1总蛋白表达水平较瘤旁组织明显增多(P=0.006);高级别胶质瘤肿瘤组织中p-TBK1蛋白水平较瘤旁明显升高,且p-TBK1的表达与CD68阳性巨噬细胞有共定位;高级别胶质瘤患者术后外周血经典单核细胞中p-TBK1表达较术前明显降低(P=0.001).结论TBK1在高级别胶质瘤微环境髓样细胞中高表达,提示髓样细胞TBK1在胶质瘤的发展中可能起着关键调控作用.
Objective To investigate the expression characteristics of TANK binding kinase 1(TBK1)in myeloid cells in the tumor microenvironment of high-grade glioma.Methods A total of 27 patients with high-grade glioma admitted to the Department of Neurosurgery,the Second Hospital of Hebei Medical University from April 2019 to February 2021 were enrolled in this study.Magnetic activated cell sorting was used to detect the expression of TBK1 in CD11b-positive myeloid cells from paratumoral and tumor tissues.The expression of p-TBK1 in macrophages in the tumor microenvironment was observed by immunofluorescence staining,and the expression of p-TBK1 in classical monocytes of peripheral blood from patients before and after surgery was analyzed by flow cytometry.Results The level of total TBK1 protein in CD1lb-positive cells of high-grade glioma was significantly increased compared with that in adjacent tissues(P=0.006).The level of p-TBK1 protein in the tumor tissue of high-grade glioma was significantly higher than that in the paratumoral tissue,and the expression of p-TBK1 was observed colocalized with CD68-positive macrophages.The expression of p-TBK1 in peripheral blood classical monocytes of patients with high-grade glioma after surgery was significantly lower than that before surgery(P=0.001).Conclusion TBK1 is highly expressed in myeloid cells in the tumor microenvironment of high-grade glioma,suggesting that myeloid TBK1 may play a key role in the development of glioma.
