基于质量常数评价方法的艾叶饮片等级评价研究

摘要 目的采用中药质量常数等级评价法对艾叶饮片进行等级划分。方法将传统外观性状与内在成分相结合,分别测定13批不同来源艾叶饮片的出绒量及指标成分含量,通过质量常数公式计算各样本的相对质量常数及百分质量常数,并以此进行等级划分。为了验证等级划分的合理性,利用脂多糖(lipopolysaccharide,LPS)诱导的巨噬细胞RAW264.7炎症模型,比较各等级艾叶的水提物及挥发油对一氧化氮(nitric oxide,NO)的抑制作用。结果所收集的13批艾叶饮片中有3批含量不符合2020版《中华人民共和国药典》规定,剩余10批合格饮片中,一等艾叶的相对质量常数≥5.19,共5批;二等艾叶的相对质量常数≥3.24且<5.19,共3批;三等艾叶的相对质量常数<3.24,共2批。抗炎结果表明,艾叶的药效作用随等级的升高而增强。结论中药质量常数等级评价法用于艾叶饮片的等级划分,具有科学性与合理性,可为艾叶饮片等级标准的建立提供依据。 Objective The quality constant grade evaluation method of traditional Chinese medicine was used to evaluate the grade of Artemisiae Argyi.Methods Combining the traditional appearance characteristics with the intrinsic components,the moxa yield and index component contents of 13 batches of Artemisiae Argyi from different sources were determined.The relative quality constant and percentage quality constant of each Artemisiae Argyi sample were calculated by the quality constant formula,and then graded.In order to verify the rationality of the results,the NO inhibitory effects of water extract and essential oil of different grades of Artemisiae Argyi were compared by using lipopolysaccharide-induced inflammation model of RAW264.7 cell.Results Among the 13 batches of Artemisiae Argyi collected,3 batches did not meet the provisions of the 2020 edition of Chinese Pharmacopoeia.And in the remaining 10 batches,five batches with the relative quality constant≥5.19 were classified as the first grade;three batches with the relative quality constant≥3.24 and<5.19 were classified as the second grade.The rest two batches were third grade,with the relative quality constant<3.24.In addition,the anti-inflammatory results showed that the efficacy of Artemisiae Argyi was enhanced with the increase of grade.Conclusion The quality constant method was scientific and reasonable for grading Artemisiae Argyi,which can provide a basis for the formulation of the grade standard of Artemisiae Argyi.
