摘要
【目的】以旱冬瓜种质资源库中定植的150棵旱冬瓜优树子代样株的11个表型性状测定值为原始数据来源,探讨旱冬瓜优质核心种质构建的适宜策略,并获得包含一定数量样株的旱冬瓜核心种质库,为旱冬瓜优质种质资源的保存、评价和利用提供科技支撑。【方法】利用单因素试验设计,采用3种遗传距离、6种系统聚类方法、4种抽样方法和6个抽样比例构建旱冬瓜种质子集,利用方差差异百分率、表型保留比例、极差符合率、Shannon-Weaver多样性指数、变异系数变化率和均值差异百分率评价不同的种质子集,通过主成分分析对所构建的旱冬瓜优质核心种质进行确认。【结果】在3种不同遗传距离构建的种质子集中,切比雪夫距离优于欧式距离和明科夫斯基距离;6种系统聚类方法构建的种质子集中,重心法优于其他5种方法;4种抽样方法构建的种质子集中,改进的最小距离逐步取样法优于多次聚类优先取样法、多次聚类偏离度取样法、多次聚类随机取样法;6种抽样比例构建的核心种质中,25%抽样比例下构建的核心种质更能充分代表原种质集。【结论】综合考虑核心种质的有效性、实用性、费用和工作量等因素,认为基于切比雪夫距离,利用重心法进行系统聚类,采用改进的最小距离逐步取样法,在25%抽样比例下构建的核心种质能充分代表原种质集,是构建旱冬瓜优质核心种质的最佳策略。
【Objective】The study used the measured values of 11 phenotypic traits of 150 offspring samples from superior Alnus nepalensis trees planted in the A.nepalensis germplasm resource library as the source of raw data.The study aimed to explore suitable strategies for constructing the A.nepalensis core germplasm of high quality,and to obtain the A.nepalensis core germplasm library containing a certain number of samples.This would provide technological support for the preservation,evaluation,and utilization of high-quality A.nepalensis germplasm resources.【Method】Using the single factor experimental design,and three genetic distances,six systematic clustering methods,four sampling methods and six sampling ratios were used to construct the proton set of A.nepalensis.The percentage of variance difference,phenotypic retention ratio,coincidence rate of range,Shannon-Weaver diversity index,percentage of variance and percentage of mean variance were used to evaluate various proton sets of A.nepalensis.The high-quality core collection of A.nepalensis was confirmed by principal component analysis.【Result】Chebyshev distance was better than European distance and Minkowski distance in the construction of high-quality core germplasm of A.nepalensis based on three genetic distance;Among six systematic clustering methods,the center of gravity method was the best;The improved minimum distance stepwise sampling method was better than priority sampling method,deviation sampling method and random sampling method;Among the six sampling ratios,the core collection constructed under 25% sampling ratio could more fully represent the original collection.【Conclusion】Comprehensively considering the effectiveness,practicability,cost and workload of core germplasm,it was considered that the core germplasm constructed by using the center of gravity method based on Chebyshev distance and the improved minimum distance stepwise sampling method under the sampling proportion of 25% could fully represent the original seed quality set,whi
