淫羊藿多糖对牛冷冻精液的影响 被引量:1

Effects of Epimedium polysaccharides on frozen semen of cattle

摘要 该试验以2kg淫羊藿为原料,先后用水提醇沉法、sevage法和阴离子交换柱提取淫羊藿多糖;取10头西蒙塔尔种牛精液,每份精液称重测量体积,用移液枪均分为5等份,用超纯水配制淫羊藿多糖溶液后稀释成品牛冷冻精液稀释液,多糖浓度分别为8、4、2mg/mL,对照组不加淫羊藿多糖,封装冷冻后检测冻精活力和畸形率;制作含有淫羊藿多糖稀释液的牛冻精进行人工授精,对比多糖冻精与普通冻精受胎率差异。结果表明:水提醇沉法得到含糖量为58.86%的淫羊藿粗多糖255.9134g,sevage法和阴离子交换柱去除蛋白质后得含糖量为94.8%淫羊藿多糖51.2411g;淫羊藿多糖在配比浓度为2mg/mL时,冻精解冻活力为(51±3.7)%、畸形率为(21.15±9.34)%,受胎率为80.00%,优于其他组。结论:94.8%淫羊藿多糖配比浓度在2mg/mL时,可最大限度保存牛精子生殖机能,人工授精受胎率最高。 In this experiment,2kg Epimedium was used as raw material,and Epimedium polysaccharide was extracted by water extraction and alcohol precipitation method,sevage method and anion exchange column.The semen of 10 Simmental cattle was weighed and measured.Each semen was divided into 5 equal parts by a pipette.The Epimedium polysaccharide solution was diluted with ultrapure water to dilute the finished bovine frozen semen diluent.The polysaccharide concentrations were 8,4,and 2 mg/mL,respectively.The control group did not add Epimedium polysaccharide,and the frozen semen vitality and deformity rate were detected after encapsulation and freezing.Bovine frozen semen containing Epimedium polysaccharide diluent was prepared for artificial insemination,and the difference of conception rate between polysaccharide frozen semen and ordinary frozen semen was compared.The results showed that 255.9134 g of crude polysaccharide with a sugar content of 58.86% was obtained by water extraction and alcohol precipitation,and 51.2411 g of polysaccharide with a sugar content of 94.8% was obtained by sevage method and anion exchange column after removing protein.When the concentration of Epimedium polysaccharide was 2 mg/mL,the thawing activity of frozen semen was(51±3.7)%,the deformity rate was(21.15±9.34)%,and the conception rate was 80.00%,which was better than other groups.Conclusion:When the concentration of 94.8% Epimedium polysaccharide is 2 mg/mL,the reproductive function of bovine sperm can be preserved to the maximum extent,and the conception rate of artificial insemination is the highest.
