摘要
目的观察肺结核患者外周血中单核细胞的活化程度与p21活化激酶5(PAK5)的表达水平,探讨单核细胞活化与PAK5的相关性。方法选择活动性肺结核患者(结核组)65例和体检健康者(对照组)65例,流式细胞术检测外周血单核细胞比例、单核细胞/淋巴细胞比值、活化单核细胞比例和数量、PAK5蛋白表达阳性率,荧光定量PCR检测PAK5基因mRNA水平,比较两组活化单核细胞中的PAK5表达差异。结果结核组外周血中单核细胞比例11.1%±2.3%高于对照组比例5.3%±1.4%,单核细胞/淋巴细胞比值(0.49±0.20)高于对照组比值(0.16±0.05),差异均有统计学意义(P<0.001)。结核组单核细胞活化比93.9%±4.6%高于对照组活化比72.9%±15.2%,单核细胞活化数(970±334)个/L高于对照组活化数(281±149)个/L,差异均有统计学意义(P<0.001)。结核组活化单核细胞PAK5蛋白表达阳性率84.71%±11.15%高于对照组阳性率75.33%±11.64%,差异均有统计学意义(P<0.001)。结核组活化单核细胞PAK5基因mRNA表达水平(0.97±0.06)高于对照组表达水平(0.16±0.07),差异均有统计学意义(P<0.001)。结论活动性肺结核患者外周血中的单核细胞活化与PAK5蛋白表达以及PAK5基因mRNA表达有关,PAK5蛋白能够促进单核细胞活化。
Objective To observe the degree of monocyte activation and the expression level of p21-activated kinase 5(PAK5)in the peripheral blood of patients with pulmonary tuberculosis,and to explore the correlation between monocyte activation and PAK5.Methods 65 patients with active pulmonary tuberculosis(tuberculosis group)and 65 healthy people(control group)were collected.Flow cytometry was used to detect the proportion of peripheral blood monocytes,the ratio of monocytes/lymphocytes,the proportion and number of activated monocytes,and the positive rate of PAK5 protein expression.Fluorescence quantitative PCR was used to detect the mRNA level of PAK5 gene,and the difference in PAK5 expression in activated monocytes between the two groups were compared.Results The percentage of monocytes in the peripheral blood of the pulmonary tuberculosis group 11.1%±2.3%was higher than that of the control group 5.3%±1.4%,and the ratio of monocytes/lymphocytes(0.49±0.20)was higher than that of the control group(0.16±0.05),and these differences were statistically significant(P<0.001).The activated ratio of monocytes in the pulmonary tuberculosis group 93.9%±4.6%was higher than that in the control group 72.9%±15.2%,and the activated number of monocytes(970±334)/L was higher than the activation number of the control group(281±149)/L,which was statistically significant(P<0.001).The positive rate of PAK5 protein expression in activated monocytes in the pulmonary tuberculosis group 84.71%±11.15%was higher than that in the control group 75.33%±11.64%,and the differences were statistically significant(P<0.001).The mRNA expression level of PAK5 gene in activated monocytes of the pulmonary tuberculosis group(0.97±0.06)was higher than that in the control group(0.16±0.07),and the difference was statistically significant(P<0.001).Conclusion The activation of monocytes in the peripheral blood of patients with active pulmonary tuberculosis is related to the expression of PAK5 protein and PAK5 mRNA.PAK5 protein can promote the activation o
