摘要
目的探讨橘皮提取物(5⁃ATAN)对多发性骨髓瘤细胞U266增殖和凋亡的影响及其机制。方法用6.25μmol/L、12.5μmol/L、25μmol/L、50μmol/L、100μmol/L的5⁃ATAN处理对数生长期U266细胞24h,四甲基偶氮唑蓝(MTT)法检测细胞增殖抑制率,流式细胞术检测细胞凋亡率和线粒体膜电位,Western blotting法检测细胞色素C、BCL⁃2、BAX、pro⁃caspase 3、pro⁃caspase 9及cleaved⁃caspase 3、cleaved⁃caspase 9、PARP、Bad、p⁃Bad的表达。结果5⁃ATAN对U266细胞的增殖有抑制作用,呈剂量依赖性(P<0.05);细胞凋亡率随药物浓度的增加显著增加(P<0.05),线粒体膜电位随5⁃ATAN浓度的增加显著降低(P<0.05)。抗凋亡蛋白Bcl⁃2、p⁃Bad及pro⁃caspase 3、pro⁃caspase 9及PARP的表达逐渐降低;细胞色素C及促凋亡蛋白Bax、cleaved⁃caspase 3、cleaved⁃caspase 9、PARP(89KDa)的表达逐渐升高(P<0.05)。结论5⁃ATAN抑制多发性骨髓瘤的增殖,促进其凋亡,可能与线粒体凋亡信号通路的激活有关。
Objective To explore whether 5⁃ATAN could induce U266 cell lines apoptosis.Methods Use U266 which is a type of multiple myeloma cell lines as the research object.MTT assay was used to detect the anti⁃growth of 5⁃ATAN.Apoptosis and mitochondrial membrane potential(MMP)were detected by flow⁃cytometry.The expression levels of BCL⁃2,BAX,pro⁃caspase 3,pro⁃caspase 9及cleaved⁃caspase 3,cleaved⁃caspase 9,PARP,Bad,p⁃Bad were detected by western blotting.Results 1.5⁃ATAN can suppress the growth of U266 cell at a dose dependent manner,the highest inhibition ratio was 80.6%which was significantly higher than the control group.The apoptotic rate in the 5⁃ATAN groups was higher than the control group(P<0.05).The mitochondrial membrane poten⁃tial is decreased with the increase concentration of 5⁃ATAN.The expression levels of Bcl⁃2,p⁃Bad,pro⁃caspase 3,pro⁃caspase 9 and PARP were decreased,while the cytochrome C,Bax,cleaved⁃caspase 3,cleaved⁃caspase 9,PARP(89KDa)were increased(P<0.05).Conclusion Treatment with 5⁃ATAN promoted apoptosis in U266 cells and inhibited their proliferation by activating the mitochondrial apoptosis signal pathway.
