摘要
目的:研究长链非编码RNA(LncRNA)HULC在结直肠癌(CRC)组织和癌旁组织中的表达,及其与CRC患者临床特征的相关性,评价其作为CRC诊断标志物的可能性。方法:收集2018-01-01至2020-01-31于我院行CRC根治术且经病理确诊的50例原发性CRC患者癌及癌旁正常组织(距癌周≥5 cm)。通过实时荧光定量聚合酶链反应(qRT-PCR)分析50对相匹配的CRC组织和癌旁组织中HULC的表达。通过配对t检验分析CRC组织和癌旁组织之间的差异性。进行单因素ANOVA方法分析LncRNA HULC的表达与患者临床病理特征的关系,通过建立ROC曲线以评估LncRNA HULC用于CRC诊断的价值。结果:HULC在CRC组织中的ΔCT值(6.59±2.26)较癌旁组织中ΔCT值(9.04±2.60)低,说明HULC在癌组织中的真正表达高于癌旁组织,差异有统计学意义(P<0.001)。癌组织中HULC的表达与CRC的分化程度(P=0.014)、淋巴结转移(P=0.007)、TNM分期(P=0.027)相关,差异有统计学意义。HULC在CRC组织及癌旁组织中表达的ROC曲线下面积为0.762,P<0.001,差异有统计学意义。结论:HULC在结直肠癌组织中呈过表达,且癌组织中HULC的表达与CRC的分化程度、TNM分期、淋巴结转移有关,对CRC的诊断可能有一定的参考价值。
Objective:To investigate the expression of long non-coding RNA(LncRNA)HULC in CRC tissues and paracancerous tissues,and its correlation with the clinical characteristics of CRC patients,and to evaluate its possibility as a diagnostic marker of CRC.Methods:50 patients with primary CRC who underwent radical CRC surgery and were pathologically confirmed in our hospital from January 2018 to January 2020 were collected for their cancer and adjacent tissues(≥5 cm from cancer circumference).The expression of hsa_circ_0079656 in matched CRC and adjacent tissues was analyzed using quantitative real-time fluorescent polymerase chain reaction(qRT-PCR).Paired t test was used to address the difference between CRC and adjacent tissues.One-way ANOVA was used to analyze the relationship between hsa_circ_0079656 expression and the clinicopathologic features of patients,and the ROC curve was established to evaluate the application value of hsa_circ_0079656 in the diagnosis of CRC.Results:TheΔCT value(6.59±2.26)of HULC in CRC tissue is lower than that ofΔCT value(9.04±2.60)in paracancer tissue,indicating that the true expression of HULC in cancer tissue is higher than that in paracancer tissue,the difference is statistically significant(P<0.001).The expression of HULC in cancer tissues was correlated with the differentiation degree of CRC(P=0.014),lymph node metastasis(P=0.007)and TNM stage(P=0.027),and the difference was statistically significant.The area under ROC curve of HULC expression in CRC and paracancer tissues was 0.762,P<0.001,the difference was statistically significant.Conclusion:HULC is overexpressed in colorectal cancer tissues,and the expression of HULC in cancer tissues is related to the differentiation degree of CRC,TNM stage and lymph node metastasis,which may have certain reference value for the diagnosis of CRC.
