摘要
目的 探讨结直肠癌组织微小RNA-16-5p(miR-16-5p)、microRNA-493-5p(miR-493-5p)表达与磷脂酰肌醇3-激酶/蛋白激酶B/哺乳动物雷帕霉素靶蛋白(PI3K/Akt/mTOR)信号通路和预后的关系。方法 选取2016年1月至2019年1月该院收治的126例接受根治性切除手术的结直肠癌患者,采用实时荧光定量聚合酶链反应(qPCR)检测癌组织及癌旁组织中miR-16-5p、miR-493-5p及PI3K/Akt/mTOR信号通路相关基因mRNA的表达水平并进行比较,分析癌组织中miR-16-5p、miR-493-5p与PI3K/Akt/mTOR信号通路相关基因mRNA及临床病理特征的关系。术后随访3年,采用Kaplan-Meier生存曲线分析miR-16-5p、miR-493-5p高表达和低表达患者的预后情况。结果 总RNA A260/A280为1.90,琼脂糖凝胶电泳提示28S与18S rRNA带亮度比值均>1.5,总RNA浓度750 ng/μL,说明RNA纯度高、完整性较好。癌组织中miR-16-5p、miR-493-5p表达水平均低于癌旁组织,PI3K、Akt、mTOR mRNA表达水平均高于癌旁组织(P<0.05)。癌组织中miR-16-5p、miR-493-5p与PI3K、Akt、mTOR mRNA表达水平均呈负相关(P<0.05)。临床分期为Ⅲ期患者中miR-16-5p、miR-493-5p低表达者占比高于Ⅰ期和Ⅱ期(P<0.05);低分化患者中miR-16-5p、miR-493-5p低表达患者占比高于中分化和高分化,且中分化患者中miR-16-5p、miR-493-5p低表达患者占比高于高分化(P<0.05);有淋巴结转移患者中miR-16-5p、miR-493-5p低表达患者占比高于无淋巴结转移(P<0.05);T4分期患者中miR-16-5p、miR-493-5p低表达患者占比高于T1和T2分期(P<0.05),T3分期患者中miR-493-5p低表达患者占比高于T1分期(P<0.05)。Kaplan-Meier生存曲线分析显示,miR-16-5p高表达和低表达患者3年累积生存率为93.22%、68.33%,生存率比较差异有统计学意义(P=0.017)。miR-493-5p高表达和低表达患者3年累积生存率为94.74%、67.74%,生存率比较差异有统计学意义(P=0.012)。结论 miR-16-5p、miR-493-5p在结直肠癌组织中表达下调,且与PI3K/Akt/mTOR信号�
Objective To investigate the relationship between microRNA-16-5p(miR-16-5p)and microRNA-493-5p(miR-493-5p)expression and phosphatidylinositol 3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/Akt/mTOR)signaling pathway and prognosis in colorectal cancer tissues.Methods A total of 126 colorectal cancer patients who were admitted to the hospital who underwent radical resection from January 2016 to January 2019 were selected.The expression levels of miR-16-5p,miR-493-5p and PI3K/Akt/mTOR signaling pathway-related genes mRNA in cancer tissues and paracancerous tissues were detected and compared by quantitative real-time fluorescence PCR(qPCR).The relationship between miR-16-5p,miR-493-5p and PI3K/Akt/mTOR signaling pathway-related gene mRNA and clinicopathological features in cancer tissues were analyzed.The patients were followed up for 3 years after operation,Kaplan-Meier survival curve was used to analyze the prognosis of patients with high expression of miR-16-5p and miR-493-5p and low expression of miR-16-5p and miR-493-5p.Results The total RNA A 260/A 280 was 1.90.Agarose gel electrophoresis showed that the brightness ratio of 28S and 18S rRNA bands was>1.5,and the total RNA concentration was 750 ng/μL,which indicated that RNA had high purity and good integrity.The expression levels of miR-16-5p and miR-493-5p in the cancer tissues were lower than those in the paracancerous tissues,and the expression levels of PI3K,Akt and mTOR mRNA were higher than those in the paracancerous tissues(P<0.05).The proportion of patients with lymph node metastasis was higher than that without lymph node metastasis(P<0.05).The proportion of patients with low expression of miR-16-5p and miR-493-5p in patients with clinical stageⅢwas higher than those in patients with stageⅠandⅡ(P<0.05).The proportion of patients with low expression of miR-16-5p and miR-493-5p in patients with low differentiation was higher than those in patients with medium differentiation and high differentiation(P<0.05),and the proportion of pati
