摘要
[目的]揭示小龙虾肠道真菌的相对丰度,分离鉴定肠道中产木聚糖酶真菌。[方法]用纯培养法从小龙虾肠道筛选产木聚糖酶真菌,对小龙虾肠道真菌进行Illumina MiSeq高通量测序,确定肠道菌群结构。[结果]从小龙虾肠道中筛选到产木聚糖酶真菌3株,菌株P3产木聚糖酶活性最高,ITS-rDNA分子鉴定显示,P3为斜卧青霉。Illumina MiSeq高通量测序结果表明:在属水平上,小龙虾肠道真菌主要是古根菌属、帚枝霉属、双足囊菌属、马拉色氏霉菌属、念珠菌属、曲霉属、Cutaneotrichosporon属、被孢霉菌属及其它未知菌属真菌。[结论]小龙虾肠道内真菌资源丰富,具有产木聚糖酶真菌-斜卧青霉。
[Objective]To reveal the relative abundance of intestinal fungi in crayfish and isolate xylanase-producing fungi.[Method]The xylanase production fungi were isolated from crayfish intestine by plate screening method,and the diversity of intestine fungi was studied by Illumina MiSeq high-throughput sequencing.[Result]Three strains of xylanase-producing fungi were screened from crayfish intestines,and strain P3 had the highest xylanase activity,ITS-rDNA molecular identification showed that strain P3 was Penicillium decumbens.The results of high-throughput sequencing showed that:at the genus level,the intestinal fungi of crayfish were mainly Archaeorhizomyces,Sarocladium,Dipodascus,Malassezia,Candida,Aspergillus,Cutaneotrichosporon,Mortierella.[Conclusion]There are abundant fungal resources in the intestine of crayfish,and there was a xylanase-producing fungus,Penicillium decumbens.
