槲皮素通过Nrf2/HO-1通路改善压力负荷诱导的心力衰竭 被引量:1

Protective effect and mechanism of quercetin on pressure overload heart failure in mice

摘要 目的 探究槲皮素(QD)对小鼠压力负荷性心力衰竭(HF)的保护作用及机制。方法 将30只健康雄性C57BL/6小鼠随机分成假手术组(Sham)、心力衰竭组(HF)和HF+QD组。采用主动脉弓缩窄(TAC)方法建立小鼠压力负荷性HF,术后给予QD腹腔注射给药,每2日1次,连续4周。收集小鼠血清及心脏组织,称取小鼠体重及心脏重量,计算心脏重量指数;HE染色观察组织病理学改变;ELISA检测小鼠血清TNF-α、IL-4和IL-10表达水平;Western blot检测小鼠心肌组织Nrf2、HO-1与COX2的蛋白表达。结果 QD能降低HF小鼠心脏重量和心脏重量指数,减轻心肌间质的炎症细胞浸润;QD能降低HF小鼠血清内TNF-α、IL-4的表达,上调IL-10的表达(P<0.05);QD能上调HF小鼠Nrf2及HO-1的蛋白表达,抑制COX2蛋白的表达(P<0.05)。结论 QD可能通过激活Nrf2/HO-1信号通路对小鼠压力负荷性HF提供保护作用。 Objective To investigate the protective effect of quercetin(QD)on pressure overload heart failure(HF)in mice.Methods Thirty healthy male C57BL/6 mice were randomly divided into sham group(Sham),heart failure group(HF)and HF+QD group.Aortic arch constriction(TAC)was used to establish pressure-loaded HF in mice.QD was given once every two days for 4 weeks after model established.Serum and heart tissue of mice were collected,body weight and heart weight of mice were measured,and heart weight index was calculated.The histopathological changes were observed by HE staining.The levels of TNF-α,IL-4 and IL-10 in serum were detected by ELISA.The expressions of Nrf2,HO-1 and COX2 in myocardial tissue of mice were detected by Western blot.Results QD can reduce the heart weight and heart weight index of HF mice,and reduce the infiltration of inflammatory cells in myocardial interstitial.QD could decrease the levels of TNF-αand IL-4 in serum of HF mice,and up-regulate the level of IL-10(P<0.05).QD could up-regulate the expressions of Nrf2 and HO-1 in HF mice,and inhibit the expression of COX2(P<0.05).ConclusioanQDmayprovide a protective effect against pressure-loaded HF in mice by activating Nrf2/HO-1 signaling pathway.
