摘要
目的:摩罗丹成为国内外治疗慢性萎缩性胃炎(Chronic Atrophic Gastritis,CAG)的推荐用药,本研究拟从网络药理学角度探讨摩罗丹治疗CAG的可能分子机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)、中医药综合数据库(TCMID)和化合物靶点预测网站等获取摩罗丹的有效成分和相关靶标蛋白,OMIM、GeneCards、DrugBank等数据库获取CAG的已知疾病靶点,经Uniprot转换后导入Cytoscape构建“药物-成分-靶点”网络,STRING数据库构建蛋白质-蛋白质相互作用(Protein-protein Interaction,PPI)网络获得核心靶标基因,基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析获得潜在作用通路。结果:(1)获得摩罗丹中有效化学成分151个,药物作用靶点760个,关键成分有豆甾醇、3-去甲基秋水仙碱、玄参苷A和芍药苷等;(2)PPI网络分析结果显示,共有73个靶点,1099个相互作用关系;(3)GO分析结果显示,摩罗丹可能通过对核因子-κB(Nuclear Factor-κB,NF-κB)和丝裂原活化蛋白激酶活性的正性调控发挥治疗CAG的作用;(4)KEGG通路富集分析结果显示,摩罗丹可能通过作用于肿瘤坏死因子(Tumor Necrosis Factor,TNF)、Toll样受体、ErbB等信号通路发挥疗效。结论:本研究初步揭示了摩罗丹通过多成分、多靶点、多通路治疗CAG。
Objective:Moluo Dan(摩罗丹)has become a recommended drug for the treatment of chronic atrophic gastritis(CAG)at home and abroad.This study aims to explore the possible molecular mechanism of Moluo Dan in the treatment of CAG from the perspective of network pharmacology.Methods:The active components and related target proteins of Moluo Dan were obtained through TCMSP,Traditional Chinese Medicine Integrative Database(TCMID)and compound target prediction website,and the known disease targets of CAG were obtained from OMIM,GeneCards,DrugBank and other databases.After the transformation by Uniprot into Cytoscape,the drug-composition-target network was constructed,the protein-protein interaction network was constructed by STRING database to obtain the core target genes,and the potential action pathway was obtained by GO and KEGG enrichment analysis.Result:①There were 151 effective chemical components in Moluo Dan and 760 drug targets,and the key components were stigmasterol,3-demethylcolchicine,scrophaloside A,paeoniflorin,etc..②PPI analysis showed that there were 73 targets and 1099 relationships in Moluo Dan.③GO analysis showed that Moluo Dan may play a role in the treatment of chronic atrophic gastritis through positive regulation of NF-κB and MAPK.④KEGG pathway enrichment analysis showed that Moluo Dan active components may exert therapeutic effects by acting on TNF signaling pathway,Toll-like receptor signaling pathway,ErbB signaling pathway and other pathways.Conclusion:This study preliminarily reveals that Moluo Dan could treat chronic atrophic gastritis with multiple components,multiple targets and multiple pathways.
