摘要
该研究采用MiSeq测序技术对采集自湖南张家界地区的18个鲊广椒样品进行高通量测序,同时从MGRAST数据库中下载12个湖南湘西地区鲊广椒的高通量测序数据,在将数据合并分析的基础上,通过多种数据方法对张家界和湘西2个地区鲊广椒菌群结构进行比较分析,探究不同地区鲊广椒细菌群落结构的异同。结果表明,2个地区鲊广椒中细菌类群丰度和多样性均无显著差异(P>0.05),但其细菌类群结构存在显著差异(P<0.05)。Mann-Whitney检验发现,张家界地区鲊广椒中乳酸杆菌(Lactobacillus)和假单胞菌属(Pseudomonas)相对含量分别为93.04%和1.78%,较之湘西地区显著偏高(P<0.05),而醋酸杆菌(Acetobacter)相对含量仅为0.03%,较之湘西地区显著偏低(P<0.05)。线性判别分析(LDA)结果表明,乳酸杆菌(Lactobacillus)和假单胞菌属(Pseudomonas)可作为张家界地区鲊广椒中的生物标志物,醋酸杆菌(Acetobacter)为湘西地区鲊广椒中的生物标志物。PICRUSt分析发现,代谢途径E(氨基酸转运与代谢)和K(转录)在张家界地区鲊广椒细菌类群中显著高表达(P<0.05)。由此可见,2个地区鲊广椒细菌类群和功能存在明显差异。
18 zha-chili samples were collected from Zhangjiajie,Hunan Province,and these samples were sequenced by MiSeq sequencing technology.At the same time,the high-throughput sequencing data of 12 zha-chili samples collected from Xiangxi,Hunan Province were downloaded from MGRAST database.The differences of bacterial groups in zha-chili samples were compared based on merge data from these two experiments by multiple data methods.The results indicated that there were no significant differences in the richness and diversity of bacterial groups among zha-chili samples collected from 2 regions(P>0.05),but there were obvious differences in bacterial group structure(P<0.05).The results of Mann-Whitney tests showed that the relative contents of Lactobacillus and Pseudomonas in zha-chili in Zhangjiajie was 93.04%and 1.78%,respectively,which was significantly higher than that in Xiangxi(P<0.05),while the relative contents of Acetobacter was only 0.03%,which was significantly lower(P<0.05).The results of linear discriminant analysis(LDA)indicated that Lactobacillus and Pseudomonas could be used as biomarkers in zha-chili samples collected from Zhangjiajie region,and Acetobacter could be used as biomarkers in zha-chili samples collected from Xiangxi region.PICRUSt analysis showed that metabolic pathways E(amino acid transport and metabolism)and K(transcription)were significantly overexpressed in the bacterial group of zha-chili samples collected from Zhangjiajie region(P<0.05).Thus,there were obvious differences in bacterial groups and functions among the zha-chili samples collected from two regions.
