摘要
目的建立离子色谱法测定布洛芬原料药中2-氯丙酸根及2,2-二氯丙酸根的含量。方法采用Thermo Dionex IonPac AS18(4 mm×250 mm)色谱柱,以20 mmol·L^(-1)氢氧化钾溶液为淋洗液,流速为1.0 mL·min^(-1),进样体积为25μL;采用抑制电导检测法,抑制电流为52 mA。结果2-氯丙酸根离子在0.00486~4.86μg·mL^(-1)内,2,2-二氯丙酸根离子在0.0400~4.00μg·mL^(-1)内与峰面积呈良好的线性关系;2-氯丙酸根离子、2,2-二氯丙酸根离子平均回收率分别为104.4%、101.6%,RSD分别为0.40%、1.8%。结论本方法简便、准确、快速,适用于布洛芬原料药中2-氯丙酸根及2,2-二氯丙酸根的含量测定。
Objective To determine the content of 2-chloropropionate and 2,2-dichloropropionate in ibuprofen by ion chromatography.Methods Thermo Dionex IonPac AS18(4 mm×250 mm)column was used with the mobile phase consisting of 20 mmol·L-1 KOH at 1.0 mL·min^(-1) and the injection volume was 25μL.Suppressed conductivity detector was used with 52 mA suppression current.Results A good linearity was shown when the sample of 2-chloropropionate was within the range of 0.00486~4.86μg·mL^(-1) and 2,2-dichloropropionate within the range of 0.0400~4.00μg·mL^(-1).The average recovery of 2-chloropropionate and 2,2-dichloropropionate was 104.4%and 101.6%with RSD of 0.40%and 1.8%,respectively.Conclusion The method is convenient,accurate and fast,which can be used for the content determination of 2-chloropropionate and 2,2-dichloropropionate in ibuprofen.
