黄甲软肝颗粒对阿尔茨海默病的药效学研究

摘要 目的探讨黄甲软肝颗粒治疗阿尔茨海默病的药理作用。方法选取8只C57BL/6J小鼠作为正常对照组,40只APP/PS1小鼠随机分为模型组,多奈哌齐组(3 mg·kg^(-1)),黄甲软肝颗粒低、中、高剂量组(1.16、2.32、4.64 g·kg^(-1))。采用Y迷宫、新物体识别和Morris水迷宫测试小鼠认知功能及记忆损伤的情况,同时取材检测各小鼠脑内海马组织中Aβ、pTau/Tau、BACE、GSK3β蛋白表达的变化,通过HE染色、尼氏染色等观察神经元排列及结构变化。结果与模型组比较,黄甲软肝高剂量组自发交替比率和目标象限穿越次数显著升高。与正常对照组比较,模型组小鼠在目标象限停留时间显著下降,黄甲软肝各剂量组均显著高于模型组。与模型组比较,黄甲软肝中、高剂量组小鼠Aβ蛋白表达显著降低;各剂量组小鼠pTau/Tau蛋白表达显著降低,海马BACE、GSK3β蛋白表达差异无统计学意义,各剂量组均显著逆转了Aβ沉积显现,尼氏小体均有所增加。结论黄甲软肝颗粒具有显著抗阿尔茨海默病作用。 Objective To determine the pharmacological effect of Huangjia Ruangan granules on Alzheimer’s disease(AD).Methods Eight C57BL/6J mice were selected as the normal control group.While 40 APP/PS1 mice were randomly divided into a model group,a Donepezil group(3 mg·kg^(-1)),Huangjia Ruangan granule low,medium and high dose groups(1.16,2.32,and 4.64 g·kg^(-1)).Y-maze,new object recognition and Morris water maze were used to test the cognitive function and memory impairment of the mice.The protein expression changes of Aβ,pTau/Tau,BACE and GSK3βin the hippocampus of each mouse brain were detected.The arrangement and structural changes of neurons were observed by HE staining and Nishi staining.Results The behavioral test showed that compared with the model group,the spontaneous alternating rate and the number of target quadrant crossing were significantly increased in the high-dose group.Compared with the normal control group,the residence time in the target quadrant of the model group decreased,and the low,medium and high dose groups were significantly higher than those in the model group.Compared with the model group,Aβprotein expression of mice in the medium and high dose groups decreased;pTau/Tau protein expression decreased in each dose groups of Huangjia Ruangan granules.There were no significant differences in the expression of BACE and GSK3βin the hippocampus among all groups.Aβdeposition was significantly reversed in each dose groups of Huangjia Ruangan granules.As to histomathological changes,the arrangement and structure of neurons in the mice did not change obviously,and the Nishi bodies in each dose groups of Huangjia Ruangan granules increased as compared with the model group.Conclusion Huangjia Ruangan granules have obvious anti-AD effect.
