摘要
为探究急性温度胁迫对红耳龟Trachemys scripta elegans消化和免疫功能的影响及饲料添加维生素E(VE)的干预效应。选取健康2龄红耳龟112只,分成未添加VE组和添加VE组(250 mg·kg^(−1)),每组设室温(30℃)、低温(17℃)和高温(35℃),于胁迫0 h、6 h、12 h取血液和肠道组织,检测血清抗氧化酶和肠道消化酶活性。结果表明,未添加VE组低温下淀粉酶和脂肪酶的活性显著上升,谷胱甘肽(GSH)含量显著降低,高温下胃蛋白酶活性极显著升高,超氧化物歧化酶(SOD)活性显著升高,GSH含量显著下降;添加VE组急性温度胁迫下的SOD活性升幅减小,GSH含量升高,低温下胃蛋白酶活性显著提高,脂肪酶活性显著降低;高温下胃蛋白酶活性极显著降低,淀粉酶活性显著升高。本研究初步阐释了急性温度胁迫对红耳龟的损伤机制,同时发现饲料添加VE具有一定程度的缓解作用,为生产中应对急性温度应激探索了一条绿色可行途径。
To explore the influence of acute temperature stress and the intervention effect of vitamin E(VE)on the digestive and immunologic function of the red-eared turtle(Trachemys scripta elegans).A total of 112 healthy T.scripta elegans(2-year-old)were divided into the groups with and without VE(250 mg·kg^(−1))intervention.Each group was further divided into the room temperature control group(30℃),the acute low temperature stress group(17℃)and the acute high temperature stress group(35℃),and the latter two groups were cooled and heated within 2 h,respectively.The blood and intestinal tissues were taken at 0 h,6 h and 12 h post stress,and the activities of serum antioxidant enzymes and intestinal digestive enzymes were detected.The results showed that under acute low temperature stress,the activities of amylase and lipase were significantly increased,while the content of glutathione(GSH)was significantly decreased(P<0.05).The activity of pepsin was extremely significantly increased(P<0.01)under acute high temperature stress,the activity of superoxide dismutase(SOD)was significantly increased,while the GSH content was significantly decreased.In the VE intervention group,the increase amplitude of SOD activity was decreased and the content of GSH was increased when the young turtle was subjected to acute temperature stress.The pepsin activity of the young turtle was significantly increased and the lipase activity was significantly decreased at 17℃.The pepsin activity was extremely significantly decreased and the amylase activity was significantly increased at 35℃.In this study,the damage mechanism of acute temperature stress on T.scripta elegans was preliminarily explained,and it was found that adding VE to feed had a certain degree of relief effect,which explored a green and feasible way to deal with acute temperature stress in production.
