摘要
目的 探讨tribbles同源物1(TRIB1)在前列腺癌(PCa)组织中的表达及与患者临床特征的关系。方法 收集92例PCa患者的PCa组织和癌旁组织,采用免疫组化染色法检测TRIB1蛋白表达情况,采用逆转录聚合酶链反应(PCR)检测TRIB1 mRNA相对表达量。比较PCa组织和癌旁组织中TRIB1蛋白表达情况及TRIB1mRNA相对表达量,比较不同临床特征PCa患者PCa组织中TRIB1 mRNA表达情况,采用Kendall相关分析法分析TRIB1 mRNA表达与PCa患者临床特征的相关性。结果 PCa组织中TRIB1蛋白阳性表达率明显高于癌旁组织(P﹤0.01)。PCa组织中TRIB1 mRNA相对表达量明显高于癌旁组织(P﹤0.01)。92例患者中,TRIB1 mRNA高表达53例,TRIB1 mRNA低表达39例。T3期、前列腺特异性抗原(PSA)水平﹥20 ng/ml、高转移负荷、Gleason评分为8~9分PCa患者PCa组织中TRIB1 mRNA高表达率分别高于T1~2期、PSA水平≤20 ng/ml、低转移负荷、Gleason评分≤7分的患者,差异均有统计学意义(P﹤0.05)。TRIB1 mRNA表达与PCa患者的转移负荷无相关性(P﹥0.05),TRIB1 mRNA表达与PCa患者的T分期、PSA水平以及Gleason评分均呈正相关(P﹤0.05)。结论 TRIB1可能参与PCa的发病过程,同时PCa的恶性程度与TRIB1 mRNA表达水平有关,研究TRIB1 mRNA表达水平对PCa的诊断和临床治疗均具有重要意义。
Objective To investigate the expression of tribbles homolog 1(TRIB1) in prostate cancer(PCa) tissues and its relationship with clinical characteristics of patients.Method PCa tissues and paracancerous tissues of 92 PCa patients were collected.The expression of TRIB1 protein was detected by the immunohistochemical staining assay.The relative expression of TRIB1 mRNA was detected by reverse transcription polymerase chain reaction(PCR).The expression of TRIB1 protein and the relative expression of TRIB1 mRNA in PCa tissues and adjacent tissues were compared.The expressions of TRIB1 mRNA in PCa tissues of PCa patients with different clinical characteristics were compared.The correlation between the expression of TRIB1 mRNA and clinical characteristics of PCa patients were analyzed by Kendall correlation analysis.Result The positive expression rate of TRIB1 protein in PCa tissues was significantly higher than that in paracancerous tissues(P<0.01).The relative expression of TRIB1 mRNA in PCa tissues was significantly higher than that in paracancerous tissues(P<0.01).Among the 92 patients,53 cases had high expression of TRIB1 mRNA,and 39 cases had low expression of TRIB1 mRNA.The high expressin rate of TRIB1 mRNA in PCa tissues of PCa patients with T3stage,prostate specific antigen(PSA) level20 ng/ml,high metastatic burden,Gleason score of 8-9 were higher than those of patients with T1-2stage,PSA level≤ 20 ng/ml,low metastatic load,and Gleason score≤ 7,and the differences were statistically significant(P<0.05).The expression of TRIB1 mRNA was not correlated with the metastatic burden of PCa patients(P<0.05),and the expression of TRIB1 mRNA was positively correlated with T stage,PSA level,and Gleason score of PCa patients(P<0.05).Conclusion TRIB1 may be involved in the pathogenesis of PCa,and the degree of malignancy of PCa is related to the expression level of TRIB1 mRNA.It is of great significance to study the expression level of TRIB1 mRNA for the diagnosis and clinical treatment of PCa.
