铁皮石斛低聚糖分离纯化及其益生活性 被引量:1

摘要 为了探究铁皮石斛低聚糖组成及其益生活性,利用DEAE-52阴离子纤维素和Sephadex G-10葡聚糖凝胶色谱柱分离纯化得到铁皮石斛低聚糖DOO1-1组分,评价了该低聚糖对干酪乳杆菌、嗜酸乳杆菌、两歧双歧杆菌和肠道菌群的益生作用。结果表明,铁皮石斛低聚糖DOO1-1含量为81.37%,其相对分子质量为2270,主要由葡萄糖和甘露糖组成,还有少量葡萄糖醛酸、半乳糖和阿拉伯糖,DOO1-1具有较强的抗消化能力。铁皮石斛低聚糖DOO1-1能促进干酪乳杆菌、嗜酸乳杆菌、两歧双歧杆菌和肠道菌群等微生物的增殖,其效果好于低聚果糖。DOO1-1能被这些益生菌利用产生短链脂肪酸,其中乳酸和乙酸含量较多,而丙酸和丁酸含量较少。试验结果表明,铁皮石斛低聚糖DOO1-1是一种良好的益生元,具有潜在的开发价值。 In order to explore the composition of Dendrobium officinale oligosaccharides and their prebiotic activity,a highly-purified oligosaccharide named DOO1-1(81.37%)from D.officinale was obtained by DEAE-52 cellulose and Sephadex G-10 gel column.An in vitro fermentation model of probiotics was established to explore the probiotic effect of DOO1-1 on Lactobacillus casei,Lactobacillus acidophilus,Bifidobacterium bifidum and intestinal bacteria.The results showed that the molecular mass of DOO1-1 was 2270 Da,which was mainly composed of glucose and mannose,with a small amount of glucuronic acid,galactose and arabinose.DOO1-1 was hardly affected by enzymes in stomach and intestine.DOO1-1 could promote the proliferation of L.casei,L.acidophilus,B.bifidum and intestinal flora,which was better than that of fructooligosaccharides.DOO1-1 could be used to produce short chain fatty acids by probiotics,with higher content of lactic acid and acetic acid,and lower content of propionic acid and butyric acid.These findings indicated that the oligosaccharide DOO1-1 from D.officinale might be a promising candidate in prebiotics.
