摘要
目的 :研究海参糖胺聚糖 (hGAG)抗血栓机制。方法 :观察hGAG对内皮细胞的促凝活性、组织因子 (TF)、凝血酶调节蛋白 (TM )和纤溶酶原激活物抑制剂 1(PAI 1)的影响。结果 :细菌脂多糖(LPS)与内皮细胞孵育后 ,不同浓度的hGAG可使细胞促凝活性下降 ,TF抗原及mRNA转录下降 ,TM抗原表达及mRNA转录增强 ;hGAG可使内皮细胞培养上清液中的PAI 1抗原、活性、mRNA转录下降。结论 :hGAG通过下调内皮细胞TF表达 ,促进TM表达 ,降低内皮细胞PAI 1合成、分泌及抑制PAI 1mRNA转录 。
AIM: To study the antithromobtic mechanism of holothurin glycoaminoglycan(hGAG). METHODS: The effects of hGAG on procoagulant activity (PCA) of endothelium, tissue factor, thrombomodulin, as well as plasminogen activitor inhibitor were observed. RESULTS: After incubation lipopolysaccharide with endothelium,the various concentrations of hGAG,could make the enhanced procoagulant activity reduced ,the increased TF:Ag level and its mRNA transcription reduced too, the expression of TM and its mRNA transcription increased. However the hGAG could make the plasminogen activator inhibitor 1(PAI-1) antigen, activity in the cell culture supernant and PAI-1 mRNA transcription significantly reduced. CONCLUSION:hGAG exerts its antithrombotic actions by means of downregulating endothelium TF expression,facilitating TM expression ,reducing PAI-1 synthesize and secretion as well as in hibiting PAI-1 mRNA transcription.
