肾细胞癌中免疫细胞浸润与患者预后的相关性分析 被引量:2

摘要 目的利用生物信息学方法,分析不同免疫细胞肿瘤浸润程度与肾细胞癌(RCC)预后的关系,为临床决策提供理论依据。方法RCC患者的基因表达谱和临床数据检索自癌症基因组图谱数据库。利用CIBERSORT算法量化免疫细胞的肿瘤浸润百分比,根据免疫浸润分数的中位数作为截断值,将患者分为各类免疫细胞的高浸润组和低浸润组。通过Kaplan-Meier曲线评估免疫细胞肿瘤浸润与患者预后的关系。利用Wilcoxon秩和检验分析免疫细胞肿瘤浸润与患者临床分期的相关性。结果树突状细胞、肥大细胞和活化型CD4+记忆性T细胞在RCC的浸润显著增加(P<0.05)。与低浸润组相比,树突状细胞和肥大细胞高浸润的RCC患者生存期显著延长(P<0.05),活化型CD4+记忆性T细胞高浸润的RCC患者生存期显著缩短(P<0.05)。此外,随着临床分期的恶性进展,树突状细胞和肥大细胞在RCC的浸润显著减少(P<0.05),活化型CD4+记忆性T细胞浸润显著增加(P<0.05)。结论免疫细胞肿瘤浸润与RCC预后相关,其中树突状细胞和肥大细胞浸润增加提示RCC预后较好,活化型CD4+记忆性T细胞浸润增加提示预后不良。 Objective To use bioinformatics methods to analyze the relationship between different degrees of immune cell infiltration in tumors and prognosis of renal cell carcinoma(RCC),so as to provide a theoretical basis for clinical decision-making.Methods Gene expression profiles and clinical data of RCC patients were retrieved from The Cancer Genome Atlas database.CIBERSORT algorithm was used to quantify the tumor infiltration percentage of immune cells.According to the median of immune infiltration score as the cut-off value,the patients were divided into high infiltration group and low infiltration group of various immune cells.Kaplan-Meier curve was used to evaluate the relationship between immune cell infiltration in tumors and patient outcomes.Wilcoxon rank sum test was used to analyze the correlation between immune cell infiltration in tumors and clinical stage of patients.Results The infiltration of dendritic cells,mast cells,and activated CD4+memory T cells in RCC was significantly increased(P<0.05).Compared with the low infiltration group,the survival of RCC patients with high infiltration of dendritic cells and mast cells was significantly prolonged(P<0.05),and the survival of RCC patients with high infiltration of activated CD4+memory T cells was significantly shortened(P<0.05).In addition,with the malignant progression of clinical stage,the infiltration of dendritic cells and mast cells in RCC significantly decreased(P<0.05),and the infiltration of activated CD4+memory T cells significantly increased(P<0.05).Conclusion The immune cell infiltration in tumors is associated with the prognosis of RCC.Increased infiltration of dendritic cells and mast cells suggests a better prognosis of RCC,and increased infiltration of activated CD4+memory T cells suggests a poor prognosis.
