摘要
目的:建立岭南有毒中草药特征性化学成分的精准识别质谱数据库及快速检测方法。方法:采用超高效液相色谱串联质谱法(UPLC-MS/MS),色谱柱为Poroshell 120 SB-C18(100 mm×2.1 mm,2.7µm),以0.1%甲酸水溶液(含5 mmol·L^(–1)甲酸铵,A)-乙腈(B)为流动相,梯度洗脱(0~1 min,10%B;1~12 min,10%~70%B;12~22 min,98%B),流速为0.3 mL·min^(–1),柱温为35℃,以三重四极杆串联质谱仪检测,离子源为电喷雾离子源(ESI),正离子模式,根据毒性成分的离子对信息、保留时间、检出限建立高通量定性定量数据库。结果:采用所建立的岭南有毒中草药精准识别质谱数据库及快速检测方法,开展毒性中草药中毒应急检验,检出钩吻、洋金花、雪上一枝蒿等毒性中草药的特征毒性成分,对粉末、药酒等不同状态、不同剂型的样本均适用。结论:所建立的分析方法快速、灵敏,筛查数据库数据准确,适合中药及中成药中毒性成分的定性定量检测,可为进一步开展中药材风险评估和药物警戒提供支撑。
Objective:To establish a precise identification mass spectrometry database and a rapid detection method for characteristic chemical components of toxic Chinese herbal medicines from the Lingnan region.Methods:Ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was employed with a Poroshell 120 SB-C18 column(100 mm×2.1 mm,2.7µm).The mobile phase consisted of 0.1%formic acid(5 mmol·L^(–1) ammonium formate)aqueous solution(A)and acetonitrile(B),using gradient elution(0-1 min,10%B;1-12 min,10%-70%B;12-22 min,98%B).The flow rate was 0.3 mL·min^(–1),and the column temperature was maintained at 35℃.Triple quadrupole tandem mass spectrometry was used with an electrospray ionization(ESI)source in the positive ion mode.A high-throughput qualitative and quantitative database was established based on ion pair information,retention time,and detection limits of toxic components.Results:The established precise identification mass spectrometry database and rapid detection method were utilized for emergency testing of toxic Chinese herbal medicines,detecting characteristic toxic components in toxic herbs such as Gelsemium elegans,Datura metel,and Artemisia argyi.The method was suitable for samples in various states and formulations,including powders and medicinal wines.Conclusion:The developed analytical method and screening database are rapid and sensitive,suitable for qualitative and quantitative detection of toxic components in Chinese medicine and herbal products,which can provide support for further risk assessment of Chinese herbal materials and pharmacovigilance.
