摘要
目的:观察金银花花蕾腺毛的形态发生及生长动态,探讨金银花花蕾腺毛生长发育与药材品质形成的相关性,为提高和稳定金银花药材质量提供新思路。方法:采集不同发育程度花蕾,分别制成石蜡切片和临时装片,观察腺毛的形态发生过程、密度、大小、颜色,并对取得数据进行统计分析。结果:金银花花蕾腺毛起源于幼嫩花蕾表皮细胞,在花蕾长约1.0 mm时开始出现,经平周与垂周分裂发育成完整腺毛;在花蕾长1.0~5.0 mm时腺毛“迅速涌现”,在花蕾长7.5 mm时腺毛密度趋于稳定;在花蕾长5.0~7.5 mm时腺毛“迅速膨大”,腺头面积、腺头周长在花蕾长10.0 mm时仍有明显增长,而腺柄面积、腺柄周长在花蕾长7.5 mm时基本不再增加;随着腺毛腺头逐渐增大,有色物质积累量增加,花蕾颜色不断加深。结论:腺毛形态发生在花蕾形态建成之后,并晚于非腺毛。腺头合成积累有色物质的趋势与腺毛发育趋势一致,提示腺毛发育与某些次生物质的合成积累密切相关,通过调控腺毛发生和发育可以调控金银花药材质量。研究结果可为金银花花蕾腺毛的深入研究提供参考。
Objective:To observe the morphogenesis and growth dynamics of glandular trichomes in Lonicera japonica Thunb.(L.japonica)buds,explore the correlation between the growth and development of glandular trichomes in L.japonica buds and the formation of the quality of medicinal materials,and provide new ideas for improving and stabilizing the quality of L.japonica medicinal materials.Methods:Flower buds with different development levels were collected and made into paraffin sections and temporary slides respectively.The morphogenesis process,density,size,and color of glandular trichomes were observed,and the data obtained were statistically analyzed.Results:The glandular trichomes in L.japonica buds originate from the epidermal cells of young buds,begin to appear when the bud is about 1 mm long,and develop into complete glandular trichomes by periclinal and anticlinal division.The glandular trichome"emerge rapidly"when the bud length is 1.0-5.0 mm,and the density of the glandular trichome tends to be stable when the bud length is 7.5 mm.When the bud length is 5.0-7.5 mm,the glandular trichome"expands rapidly",and the area and circumference of the glandular head still increase significantly when the bud length is 10.0 mm,while those of the glandular stalk basically no longer increase when the bud length is 7.5 mm.As the glandular head of the glandular trichome gradually increases,the accumulation of colored substances increases,and the color continues to deepen.Conclusion:The morphology of glandular trichomes occurs after the morphology of the flower bud is built and is later than that of non-glandular trichomes.The trend of glandular head synthesis and accumulation of colored substances is consistent with that of glandular trichome development,suggesting that glandular trichome development is closely related to the synthesis and accumulation of some secondary substances.The quality of L.japonica medicinal materials can be regulated by regulating the generation of glandular trichomes.The results of this study can provid
