小麦、野生二粒小麦、大麦CBF基因家族比较分析

摘要 CBF(C⁃repeat⁃binding factor)是植物响应低温胁迫的主要转录因子。对小麦、野生二粒小麦、大麦CBF基因家族进行了全基因组鉴定和比较分析,并采用生物信息学技术对CBF基因的结构、系统发育、染色体定位、顺式作用元件以及共线性进行了分析,然后利用转录数据分析了该基因在不同胁迫条件下以及不同组织中的表达模式。经鉴定,小麦、野生二粒小麦、大麦CBF基因分别有75、31、19个。CBF基因具有很强的保守性,编码蛋白为亲水蛋白,绝大多数具有单一外显子结构,在第5染色体上集中分布;CBF基因家族存在片段串联重复和染色体倍加现象;不同CBF基因具有不同顺式作用元件。在不同胁迫条件下CBF基因表达水平不同,不同组织中的CBF基因表达水平也不同。由此推测CBF基因结构和功能出现分化可能是植物对不同环境的自适应结果,片段串联重复和染色体倍加是CBF基因家族扩张的驱动力。 C⁃repeat⁃binding factor(CBF)is the main transcription factor in plant response to low temperature stress.In this study,the CBF gene family were identified from the whole genome of wheat,wild emmer and barley to analyze and compare their structure,phylogeny,chromosome location,cis⁃acting element and collinearity.Meanwhile,their expression patterns in different tissues under different stress conditions were analyzed by transcriptional data.The results showed that there were 75,31 and 19 CBF genes in wheat,wild emmer and barley,respectively.The CBF genes were highly conserved and centrally distributed on chromosome 5;they encoded hydrophilic protein and most of them had a single exon structure.The CBF gene family extended in the chromosome through tandem duplication and chromosomal segment duplication,and different CBF genes were composed of different cis⁃acting elements.The expression level of CBF genes varied under different stress conditions and in different tissues.It is speculated that the diversity of CBF genes in structure and function may result from the self⁃adaption of plant to different environments,and the expansion of CBF gene family is driven by tandem duplication and chromosomal segment duplication.
