基于数据挖掘分析精神分裂症不同脑区差异性基因表达及其与免疫的相关性

Analysisof diferential gene expression in different brain regions of schizophrenia and its correlation withimmunity based on Data Mining

摘要 目的 分析精神分裂症(Schizophrenia,SZ)患者在不同脑区中的差异表达基因,寻找SZ生物标志物.方法 以"Schizophrenia sequencing"为关键词,在GEO数据库检索.从该基因库中提取60例SZ患者及55例正常对照的测序结果,利用DESeq2软件计算各组差异表达基因,基于生物学过程(Biological Process,BP)采用GO功能分析,差异分析,交集分析等方法,探索SZ患者中多个脑区均异常表达基因的相关生物学过程及其与免疫的相关性,挑选调控SZ的标记基因.采用荧光定量PCR检测正常对照、SZ患者治疗前、后血液中ARG2、PLXNB3表达水平.结果 SZ在BA9、BA11、BA24脑区显著高表达的基因总共有2059个,低表达基因2002个(P<0.05),经过分析最终得到8个免疫相关候选基因,与海马CA3区相对比,发现SZ患者ARG2、BDNF、CETP、CMTM5、PLXNB3,表达下调(P<0.05),其中,ARG2、PLXNB3(P=0.018、0.024)的表达水平治疗后升高.结论 免疫相关基因可能参与SZ的调控,ARG2、PLXNB3表达水平改变与SZ的发病和治疗相关,可能作为精神分裂症的标志基因. Objective To analyze the correlation between differential molecules and immunity in Schizophrenia(SZ)patients,and screen the biomarker genes regulating SZ.Methods With Schizophrenia Sequencing as the key word,it was searched in the GEO database.The differentially expressed genes in each group were calculated by DESeq2 software.GO functional analysis,differential analysis and intersection analysis were used based on Biological Process(BP)to select marker genes regulating SZ.Fluorescent quantitative PCR was used for laboratory validation.Results There were 2059 highly expressed genes and 2002 low expressed genes in BA9,BA1l and BA24 of SZ,P<0.05.Through analysis,it was found that the expression of ARG2,BDNF,CETP,CMTM5 and PLXNB3 in SZ patients was down regulated(P<0.05),among which the expression level of ARC2 and PLXNB3(P=0.018,0.024)increased af-ter treatment.Conclusions The changes in the expression levels of ARG2 and PLXNB3 are related to the pathogenesis and treatment of SZ,which can be used as a marker gene for schizophrenia.
