摘要
目的:以干酵母发热大鼠模型为基础,研究不同时机针刺大椎时体温与cAMP的变化趋势,以探究发热大鼠的最佳针刺时机和体温降低幅度改变的机制。方法:450只SD大鼠,随机选取10只大鼠为空白对照组,其余大鼠使用20%酵母悬液进行发热模型的建立,然后随机分为模型对照组90只、上升期治疗组90只、峰值前治疗组80只、峰值治疗组70只、峰值后治疗组60只。各组进行体温与cAMP含量测定。结果:4个治疗组均可使发热大鼠体温降至空白对照组水平,解热时间:峰值治疗组(3 h)<峰值后治疗组(4 h)<峰值前治疗组(6 h)<上升期治疗组(8 h);各治疗组体温降低幅度和cAMP降低幅度由大到小依次为峰值治疗组、峰值后治疗组、峰值前治疗组、上升期治疗组;体温降低值与cAMP降低值具有显著正相关关系。结论:不同时机针刺干酵母致热大鼠大椎穴均有退热作用并可使体温降至正常水平,以体温到达峰值时进行针刺治疗效果最佳。不同时机针刺发热大鼠大椎穴可能是通过改变cAMP等中枢体温正调节介质的降低幅度来实现改变体温降低幅度。
Objective:Based on the rat model of dry yeast fever,the change trend of body temperature and cAMP during acupuncture of Dazhui(GV14)at different time was studied,so as to explore the best acupuncture time and the mechanism of changes in the reduction magnitude of body temperature decrease in febrile rats.Methods:450 SD rats were taken,from which,10 rats were randomly selected as blank control group,and the fever model was established of the remaining rats using 20%yeast suspension.The remaining rats were randomly divided into model control group(n=90),rising stage treatment group(n=90),pre-peak treatment group(n=80),peak treatment group(n=70),and post-peak treatment group(n=60).Body temperature and cAMP content were measured in each group.Results:All the 4 treatment groups could reduce the body temperature of feverish rats to the level of blank control group.The antipyretic times were:peak treatment group(3 h)<post-peak treatment group(4 h)<pre-peak treatment group(6 h)<rising stage treatment group(8 h).The decreasing amplitude of body temperature and cAMP in each treatment group was(in descending order):peak treatment group,post-peak treatment group,pre-peak treatment group,and rising stage treatment group.There is a significant positive correlation between the decrease in body temperature and the decrease in cAMP.Conclusion:Acupuncture at Dazhui(GV14)point of dry yeast induced fever in rats at different time all has antipyretic effect and can reduce the body temperature to normal level.Acupuncture at the peak of body temperature has the best effect.Acupuncture at the Dazhui point of febrile rats at different time may alter the decrease in body temperature by altering the amplitude of central temperature positive regulatory mediators such as cAMP.
