复方中药免疫增强剂对寒冷刺激小鼠呼吸道黏膜的保护作用研究 被引量:3

Study on the protective effects of compound Chinese medicine immunopotentiator on respiratory mucosa of cold-stimulated mice

摘要 为了探讨复方中药免疫增强剂对小鼠呼吸道黏膜的保护作用,试验将36只C57BL/6小鼠随机分为空白对照组、模型组和复方中药免疫增强剂组,每组12只。复方中药免疫增强剂组给药剂量为0.5 g/kg,溶于0.4 mL无菌水中灌胃给予,1次/d,连续7次;空白对照组和模型组灌胃等体积0.9%氯化钠溶液。末次给药30 min后,模型组和复方中药免疫增强剂组小鼠置-20℃环境中1 h,再转移至25℃环境中30 min,循环刺激2次以建立寒冷刺激致小鼠鼻黏膜免疫屏障功能低下模型;空白对照组小鼠不予任何刺激。采用试剂盒检测小鼠上呼吸道中分泌型免疫球蛋白A(sIgA)、λ干扰素(IFN-λ)、溶菌酶(LZM)分泌水平,观察鼻黏膜组织的病理变化,测定各组小鼠免疫器官指数。结果表明:与空白对照组比较,模型组小鼠sIgA、IFN-λ质量浓度均显著下降(P<0.05),表明寒冷刺激致小鼠鼻黏膜免疫屏障功能低下模型制备成功。与模型组比较,复方中药免疫增强剂组小鼠sIgA、IFN-λ质量浓度均显著上升(P<0.05)。空白对照组鼻黏膜上皮结构完整、纤毛分布规整,未见明显脱落,淋巴细胞丰富;模型组鼻黏膜上皮萎缩,纤毛脱落,结构紊乱;复方中药免疫增强剂组黏膜上皮结构完整,纤毛分布较规整,和模型组比较,淋巴细胞增多。与空白对照组比较,模型组小鼠LZM活性、脾脏指数和胸腺指数均显著下降(P<0.05);与模型组比较,复方中药免疫增强剂组小鼠LZM活性、脾脏指数和胸腺指数均显著上升(P<0.05)。说明复方中药免疫增强剂可通过促进sIgA、IFN-λ和LZM的分泌,提高免疫器官指数来增强小鼠鼻黏膜免疫屏障功能,维持鼻黏膜形态结构完整,改善机体的免疫功能。 In order to explore the protective effects of compound Chinese medicine immunopotentiator on the respiratory mucosa of mice,in the experiment,36 C57BL/6 mice were randomly divided into the blank control group,the model group and the compound Chinese medicine immunopotentiator group,with 12 mice in each group.The dose of compound Chinese medicine immuneopotentiator group was 0.5g/kg,which was dissolved in O.4 mL sterile water by intragastric administration,1 time/d,7 consecutive times;the blank control group and model group were given equal volume of 0.9%sodium chloride solution.After 30 min of the last administration,mice in the model group and the compound Chinese medicine immunopotentiator group were placed at-20℃for 1 h,and then transferred to 25℃for 30 min.Two times of cyclic stimulation were carried out to establish cold stimulation-induced model mice of nasal mucosal immune barrier dysfunction.The normal group of mice was not stimulated in any way.The secretion levels of secretory immunoglobulin A(slgA),λinterferon(IFN-λ)and lysozyme(LZM)in the upper respiratory tract of mice were detected by kit.The pathological changes of nasal mucosal tissues were observed,and the immune organ indexes of mice in each group were measured.The results showed that compared with the blank control group,the mass concentrations of slgA and IFN-λ in the model group decreased significantly(P<0.05),which indicated that the model with low immune barrier function of the nasal mucosa stimulated by cold was successfully prepared.Compared with the model group,the mass concentrations of slgA and IFN-λ in the compound Chinese medicine immunopotentiator group increased significantly(P<0.05).The nasal mucosal epithelium in the blank control group had a complete structure,regular cilia distribution,no obvious shedding,and abundant lymphocytes;there were nasal mucosal epithelial atrophy,cilia shedding,structural disorders in the model group;the mucosal epithelium structure in the compound Chinese medicine immunopotentiator group was
