摘要
试验旨在确定引起新疆昌吉市某规模化猪场仔猪死亡的病因,采集病死猪的脾脏、肝脏、心脏、肺脏、肾脏和淋巴结等组织样进行细菌病原分离鉴定。分离菌株进行纯化培养、革兰氏染色镜检、生化试验、菌种鉴定及测序分析、血清型鉴定和药敏试验。结果显示,分离株在TSA营养琼脂上生长良好,镜检可见革兰氏阳性球菌,呈典型链状排列;生化特性与猪链球菌相符;PCR扩增得到大小为328 bp和689 bp的目的条带,分离株与猪链球菌同源性为100%,在系统进化树上与2型猪链球菌处于同一分支,为2型猪链球菌;分离株对恩诺沙星、环丙沙星、林可霉素、阿莫西林、头孢噻呋钠、氟苯尼考6种药物敏感,对磺胺嘧啶钠耐药。研究表明,该猪场仔猪感染了猪链球菌,恩诺沙星、环丙沙星等药物可用于临床治疗。
The purpose of the experiment was to determine the cause of death of piglets in a large-scale pig farm in Changji City,Xinjiang,and to isolate and identify bacterial pathogens from spleen,liver,heart,lung,kidney and lymph nodes of dead pigs.The isolated strains were purified culture,Gram staining microscopy,biochemical test,strain identification and sequencing analysis,serotype identification and drug susceptibility test.The results showed that the isolates grew well on TSA nutrient AGAR,and Gram-positive cocci were found in typical chain arrangement.Biochemical characteristics were consistent with Streptococcus suis.The target bands with the sizes of 328 bp and 689 bp were obtained by PCR amplification.The homology of the isolates with Streptococcus suis was 100%,and the isolates were in the same branch as Streptococcus suis type 2 in the phylogenetic tree.The isolates were sensitive to enrofloxacin,ciprofloxacin,lincomycin,amoxicillin,ceftiofuran sodium and flufenicol,and resistant to sulfadiazine sodium.The study indicates that the piglets are infected with Streptococcus suis type 2.Enrofloxacin,ciprofloxacin and other drugs could be used for clinical treatment.
