定量核磁共振波谱法测定枣果实中的可溶性糖含量 被引量:1

Determination of Soluble Sugar in Jujube by Quantitative Nuclear Magnetic Resonance Spectroscopy

摘要 建立定量核磁共振波谱法测定枣果实中葡萄糖、果糖、蔗糖3种可溶性糖含量的方法。样品用氘代水超声提取,以邻苯二甲酸氢钾为内标,Zgpr作为测定序列进行氢谱测定。结果表明,葡萄糖、果糖、蔗糖线性方程斜率的测定值与理论值误差小于2%,线性相关系数均大于0.9999,精密度RSD小于0.5%,3种糖的检出限分别为23.9、20.0、18.0μg/mL,定量限分别为79.7、66.7、60.0μg/mL,回收率分别为93.0%~98.5%、92.3%~94.7%、94.9%~99.2%。对市售样品进行测定,结果与国标高效液相色谱法一致,表明该法测定枣果实中葡萄糖、果糖、蔗糖含量准确可靠,且简便、快速,可为枣原料及其加工食品的质量控制提供参考。 A quantitative nuclear magnetic resonance spectroscopy(qNMR)method was established to determine the content of soluble sugar(glucose,fructose and sucrose)in jujube.The sample was extracted with deuterated water by ultrasonic,and determined by hydrogen spectroscopy using potassium hydrogen phthalate as internal standard and Zgpr as determination sequence.The results showed that the error between the measured value and the theoretical value of the linear equation slop of glucose,fructose and sucrose was less than 2%,the linear correlation coefficients were greater than 0.9999,the RSD was less than 0.5%,the limit of detection of the three sugars were 23.9μg/mL,20.0μg/mL,18.0μg/mL,limit of quantitation were 79.7μg/mL,66.7μg/mL,60.0μg/mL,the recoveries were 93.0%-98.5%,92.3%-94.7%,94.9%-99.2%respectively.The results of the determination of commercial samples were consistent with those of the national standard high performance liquid chromatography method,which showed that the qNMR method was accurate,reliable,simple and rapid,and could provide reference for the quality control of jujube and its products.
