FOXA1在乳腺癌中的研究进展

摘要 乳腺癌的发病与家族遗传和自身雌激素水平密切相关,其发生和发展受多种转录因子调控。叉头盒蛋白A1(FOXA1)是转录调节因子FOX家族成员之一,通过与染色体结合释放出DNA结合位点,对信号转导和细胞增殖进行调控。FOXA1通过与多个基因的启动子相结合,参与并介导调控乳腺癌细胞增殖、分化、周期、侵袭、凋亡及上皮间质转化。FOXA1表达与雌激素受体、孕激素受体呈正相关,与乳腺癌化疗、内分泌治疗和靶向治疗的敏感性有关,是乳腺癌良好预后的重要预测因子之一。本文对FOXA1在乳腺癌中的研究进展进行综述,为FOXA1在乳腺癌发展中的作用机制研究及治疗提供理论依据。 The incidence of breast cancer is closely related to family genetics and its own estrogen level,and its occurrence and development are regulated by a variety of transcription factors.Fork-head box protein A1(FOXA1)is a member of the transcription regulator FOX family.It releases DNA binding sites by binding with chromosomes to regulate signal transduction and cell proliferation.FOXA1 participates in and mediates the regulation of breast cancer cell proliferation,differentiation,cycle,invasion,apoptosis and epithelial mesenchymal transformation by combining with multiple gene promoters.As one of the important predictors of good prognosis of breast cancer,the expression of FOXA1 is positively correlated with estrogen and progestin and is related to the sensitivity of chemotherapy,endocrine therapy and targeted therapy of breast cancer.This article reviews the research progress of FOXA1 in breast cancer,and provides a theoretical basis for the mechanism and treatment of FOXA1 in the development of breast cancer.
