摘要
研究超声辅助提取油茶叶中黄酮的工艺条件。通过单因素试验设计,对影响黄酮得率的料液比、乙醇体积分数、超声时间、超声温度、木瓜蛋白酶添加量及提取次数等因素进行探究,然后利用Design Expert 8.0软件进行Box-benhnken试验设计,乙醇体积分数、超声时间和木瓜蛋白酶添加量为独立变量,黄酮得率为响应值进行方差分析和多项式拟合回归,建立回归数学模型。结果表明,超声功率240 W,频率40 kHz,乙醇体积分数75%,木瓜蛋白酶添加量0.8%,料液比1∶22(g∶mL),超声温度50℃,超声时间55 min,提取的油茶叶黄酮平均得率为(2.837±0.371)%。
The technological conditions of ultrasonic assisted extraction of flavonoids from Camellia oleifera leaves were studied.Through the single factor experimental design,the factors affecting the yield of flavonoids,such as solid-liquid ratio,ethanol concentration,ultrasonic time and temperature,papain dosage and extraction times,were explored,and then the Box-be-nhnken experimental design was carried out by using Design Expert 8.0 software.The ethanol concentration,ultrasonic time and enzyme addition were independent variables,and the yield of flavonoids was the response value for variance analysis and polynomial fitting regression,and the regression mathematical model was established.The results showed that the average yield of flavonoids in Camellia oleifera leaves was(2.837±0.371)%with ultrasonic power 240 W,frequency 40 kHz,ethanol concentration 75%,enzyme addition 0.8%,solid-liquid ratio of 1∶22(g∶mL),ultrasonic treatment at 50℃for 55 min.
