摘要 目的明确circRNA_102272在肝癌中的表达及作用机制。方法通过分析GEO数据库数据以及收集锦州医科大学附属第一医院肝癌患者组织标本,通过qPCR方法检测患者组织中circRNA_102272的表达情况,通过CCK-8分析其对肝癌细胞增殖的影响,分析circRNA_102272的表达与临床病例参数之间的关系,通过在线数据库分析其作用机制,通过体外荧光素酶报告基因实验以及CCK-8明确其发挥作用的机制。结果肝癌患者组织中circRNA_102272表达较癌旁正常组织中的表达升高显著。同时,circRNA_102272的表达与分化程度、转移、TNM分期相关(P<0.05)。miR-1258能够逆转circRNA_102272对肝癌细胞增殖的调控作用。结论circRNA_102272通过miR-1258调控肝癌细胞增殖。 Objective To clarify the expression and mechanism of circRNA_102272 in liver cancer.Methods The expression of circRNA_102272 in the tissues of patients with liver cancer in the First Affiliated Hospital of Jinzhou Medical University was detected by qPCR through the analysis of GEO database data and the collection of tissue samples.The effect of circRNA_102272 on the proliferation of hepatocellular carcinoma cells was analyzed by CCK-8,and the relationship between the expression of circRNA_102272 and clinical case parameters was analyzed.The mechanism of its action was analyzed by online database,and the mechanism of its action was clarified by in vitro luciferase reporter gene assay and CCK-8.Results The expression of circRNA_102272 in liver cancer patients was significantly higher than that in adjacent normal tissues.The expression of circRNA_102272 was correlated with differentiation,metastasis and TNM staging(P<0.05).MiR-1258 could reverse the regulatory effect of circRNA_102272 on the proliferation of hepatoma cells.Conclusion CircRNA_102272 regulates the proliferation of liver cancer cells through miR-1258.
