摘要
目的:观察硫酸脑苷脂(sulfatide)活化Ⅱ型自然杀伤(NK)T细胞对小鼠肺Ⅰ型NKT细胞分泌白细胞介素(IL)-10水平的影响。方法:10只野生型BALB/c小鼠随机分为野生型sulfatide治疗组和野生型正常对照组,每组5只;10只CD1d^(-/-)BALB/c小鼠随机分为CD1d^(-/-)sulfatide治疗组和CD1d^(-/-)正常对照组,每组5只。野生型和CD1d^(-/-)sulfatide治疗组小鼠单次腹腔注射sulfatide 20μg,野生型和CD1d^(-/-)正常对照组以等量PBS代替。应用荧光定量聚合酶链反应法检测各组小鼠肺组织IL-10 mRNA表达水平;酶联免疫吸附法检测各组小鼠支气管肺泡灌洗液(BALF)中IL-10水平;流式细胞术检测各组小鼠肺Ⅰ型和Ⅱ型NKT细胞数量及IL-10+Ⅰ型和IL-10+Ⅱ型NKT细胞数量。结果:野生型sulfatide治疗组小鼠肺组织IL-10 mRNA表达水平和BALF中IL-10水平明显高于野生型正常对照组和CD1d^(-/-)sulfatide治疗组(P<0.01,P<0.001或P<0.05),但CD1d^(-/-)sulfatide治疗组小鼠肺组织IL-10 mRNA表达水平和BALF中IL-10水平与CD1d^(-/-)正常对照组比较差异无统计学意义(均P>0.05)。野生型sulfatide治疗组小鼠肺Ⅱ型NKT细胞数量明显高于野生型正常对照组(P<0.001),但两组小鼠肺Ⅰ型NKT细胞数量比较差异无统计学意义(P>0.05)。野生型sulfatide治疗组小鼠肺IL-10+Ⅰ型NKT细胞数量明显高于野生型正常对照组(P<0.001),但两组小鼠肺IL-10+Ⅱ型NKT细胞数量比较差异无统计学意义(P>0.05)。结论:硫酸脑苷脂活化Ⅱ型NKT细胞可以诱导小鼠肺Ⅰ型NKT细胞分泌IL-10。
Objective:To investigate the effects of sulfatide⁃reactive typeⅡnatural killer T cells(NKT cells)on the number of lung interleukin(IL)⁃10+typeⅠNKT cells in mice.Methods::Ten female wild⁃type(WT)BALB/c mice and ten female CD1d^(−/−)BALB/c mice were randomly divided into PBS adminis⁃tration group(n=5)and sulfatide administration group(n=5).The mice in sulfatide administration groups including WT and CD1d^(−/−)mice were intraperitoneally administrated a single injection of 20μg sulfatide,while the mice in PBS administration group were intraperitoneally injected with the same volume of PBS.The level of IL⁃10 mRNA of lung tissues was detected by using realtime fluorescence quantitative PCR;the concentration of IL⁃10 in the bronchial alveolar lavage fluid(BALF)was mea⁃sured by enzyme⁃linked immunosorbent assay,while the numbers of lung typeⅡNKT cells,typeⅠNKT cells,IL⁃10⁃producing typeⅡNKT cells and typeⅠNKT cells were analyzed by flow cytome⁃try.Results::WT mice treated with sulfatide displayed the increased expression of lung IL⁃10 mRNA and higher concentration of IL⁃10 in BALF when compared with those treated with PBS or CD1d^(−/−)mice treated with sulfatide(P<0.01,P<0.001,or P<0.05,respectively).However,no significant difference was observed in the expression of lung IL⁃10 mRNA or concentration of IL⁃10 in BALF from CD1d^(−/−)mice adminstrated with sulfatide and PBS(all P>0.05).Sulfatide treatment markedly en⁃hanced the number of lung typeⅡNKT cells(P<0.001),but not typeⅠNKT cells in the WT mice(P>0.05).However,the number of lung IL⁃10+typeⅠNKT cells was significantly increased in WT mice treated with sulfatide as compared with that treated with PBS(P<0.001).Conclusion::Sulfatide⁃reactive typeⅡNKT cells can promote lung typeⅠNKT cells to secret IL⁃10 in mice.
