摘要
目的 分析银杏叶提取物对结肠癌患者SW480细胞增殖的影响。方法 选择2022年1—12月新疆五家渠第六师总医院的10份结肠癌标本为研究对象,并对SW480细胞标本进行体外培养,然后实施100、200、300、400、500 mg/L浓度的银杏叶提取物进行反应,并通过CCK-8对人结肠癌SW480细胞的增殖抑制率进行检测,对比干预不同浓度的干预效果。结果 空白对照组的吸光度为(1.125±0.139),100、200、300、400、500 mg/L浓度银杏叶提取物处理后的吸光度分别为(1.145±0.214)、(0.943±0.018)、(0.616±0.017)、(0.571±0.039)、(0.508±0.014),200、300、400、500 mg/L浓度时的吸光度与空白对照组比较,差异有统计学意义(t=4.106、11.474、12.135、13.966,P<0.05)。空白对照组的抑制率为(0.002±0.001)%,100、200、300、400、500 mg/L浓度银杏叶提取物处理后的抑制率分别为(3.020±0.124)%、(12.500±1.242)%、(40.000±4.252)%、(43.100±4.626)%、(45.200±4.252)%,分别与空白对照组比较,差异有统计学意义(t=76.963、31.821、29.747、29.461、33.614,P<0.05)。100、200、300、400、500 mg/L浓度银杏叶提取物处理后的活性氧(reactive oxygen species,ROS)、X连锁凋亡抑制蛋白(X-linked inhibitor of apoptosis protein, XIAP)mRNA表达水平与空白对照组比较,差异有统计学意义(P<0.05)。结论 银杏叶提取物可对结肠癌SW480细胞的增殖表现为浓度依赖性地抑制。
Objective To analyze the effect of ginkgo biloba extract on the proliferation of SW480 cells in patients with colon cancer.Methods Ten colon cancer specimens from Xinjiang Wujiaqu Sixth Division General Hospital from January to December 2022 were selected as the research objects,and SW480 cell specimens were cultured in vitro.Then ginkgo biloba extract with concentrations of 100 mg/L,200 mg/L,300 mg/L,400 mg/L and 500 mg/L was admin⁃istered for reaction,and the proliferation inhibition rate of human colon cancer SW480 cells was detected by CCK-8,and the intervention effects of different concentrations were compared.Results The absorbance of the blank control group was(1.125±0.139).The absorbance of ginkgo biloba extract treated with 100 mg/L,200 mg/L,300 mg/L,400 mg/L and 500 mg/L concentration was(1.145±0.214),(0.943±0.018),(0.616±0.017),(0.571±0.039)and(0.508±0.014),re⁃spectively.Absorbance at 200 mg/L,300 mg/L,400 mg/L and 500 mg/L concentrations was compared with that of the blank control group,the difference was statistically significant(t=4.106,11.474,12.135,13.966,P<0.05).The inhibition rate of the blank control group was(0.002±0.001%).The inhibition rates of ginkgo biloba extract treated with 100 mg/L,200 mg/L,300 mg/L,400 mg/L and 500 mg/L concentration were(3.020±0.124)%,(12.500±1.242)%,(40.000±4.252)%,(43.100±4.626)%and(45.200±4.252)%,respectively,compared with the blank control group,the difference was statistically significant(t=76.963,31.821,29.747,29.461,33.614,P<0.05).The mRNA expression lev⁃els of reactive oxygen species(ROS)and X-linked inhibitor of apoptosis protein(XIAP)of ginkgo biloba extract treated with 100 mg/L,200 mg/L,300 mg/L,400 mg/L and 500 mg/L concentrations were compared with those of the blank control group,the difference was statistically significant(P<0.05).Conclusion Ginkgo biloba extract inhibited the proliferation of colon cancer SW480 cells in a concentration-dependent manner.
