摘要
为研究甘薯小孢子发育时期与花器外观形态的相关性,以冀粉2号、广薯15-156和紫罗兰为试验品种,选取不同纵径、横径的花蕾,采用醋酸洋红染色法对小孢子发育时期进行细胞学观察,确定小孢子不同发育时期及其形态特征,并对花蕾大小与小孢子发育时期的对应关系进行分析.结果表明:3个甘薯品种小孢子发育过程主要经历小孢子母细胞时期、四分体时期、单核期和成熟花粉粒期,每个发育时期特征明显;小孢子发育时期与花器外观形态密切相关,花蕾大小可以作为判断小孢子发育时期的主要依据;3个甘薯品种小孢子处于单核期时,花蕾明显变大,花萼顶端有开裂,花蕾纵径为9.30~9.41 mm,横径为2.52~2.53 mm,花药长度为1.29~1.46 mm.研究结果可为甘薯花药或小孢子培养提供细胞学依据.
To explore the correlation between the microspore development stage and the morphology of floral organ in sweetpotato,Jifen 2,Guangshu 15-156 and Violet were used as test varieties.Flower buds with different longitudinal and transverse diameters were selected.The cytological observation of microspore development stage was carried out by using Aceto-Carmine staining method to determine the different development stages of microspore and their respective morphological characteristics,and the correspondence relationship between bud size and microspore development stage was analyzed.The results showed that the microspore development of the three sweetpotato varieties mainly experienced microspore mother cell stage,tetrad stage,the uninucleate stage and mature pollen grain stage,with obvious characteristics in each development stage.The development stages of microspore were closely related to the morphology of floral organs,and the bud size could be used as the main basis for judging the development period of microspore.When the microspores of the three sweetpotato varieties were at the uninucleate stage,the flower buds became significantly larger,the top of the calyx was cracked,the longitudinal diameters of the flower buds were 9.30-9.41 mm,the transverse diameters of the flower buds were 2.52-2.53 mm,and the lengths of the anthers were 1.29-1.46 mm.The results could provide cytological basis for anther or microspore culture of sweetpotato.
