摘要
目的:观察防己水煎液及各拆分组分对阿霉素诱导的肾病综合征大鼠作用的影响,找寻防己对阿霉素肾病模型作用的药效物质基础。方法:采用尾静脉注射阿霉素的方法建立大鼠阿霉素肾病模型。第1 d、第8 d分别按4 mg/kg、3.5 mg/kg大鼠尾静脉注射阿霉素。第11 d开始灌胃给予防己水煎液1.36 g/kg、多糖组分0.82 g/kg、生物碱组分0.19 g/kg、非生物碱组分0.17 g/kg、甾体组分0.03 g/kg及阳性对照泼尼松6.3 mg/kg,连续给药14 d。检测大鼠24 h尿蛋白,血清中ALB、TP、TC、TG、SCr、BUN含量,IL-1β、TNF-α含量,MDA、GSH-Px、SOD氧化应激水平;HE染色观察大鼠肾脏病理学改变。结果:与正常对照组比较,模型对照组大鼠尿中蛋白含量显著升高,血清SCr、BUN、TC、TG、MDA含量显著升高,IL-1β、TNF-α含量显著升高(P<0.01),血清ALB、TP、GSH-Px含量、SOD活力显著降低,大鼠肾脏指数显著升高(P<0.01);与模型对照组比较,防己水煎液1.36 g/kg组、多糖0.82 g/kg组、生物碱0.19 g/kg组可明显升高血清ALB含量(P<0.05或P<0.01),降低血清TC、BUN、TNF-α、MDA含量及24 h尿中蛋白浓度(P<0.05或P<0.01),升高GSH-Px、SOD活力(P<0.01);多糖0.82 g/kg组可升高TP含量(P<0.01);水煎液1.36 g/kg组、多糖0.82 g/kg组可降低TG含量(P<0.05或P<0.01);水煎液1.36 g/kg组、生物碱0.19 g/kg组可降低SCr含量(P<0.05或P<0.01);水煎液1.36 g/kg组、多糖0.82 g/kg组可降低IL-1β含量(P<0.01)。三组大鼠肾脏病理改变明显,蛋白管型和胶样管型明显减少,肾曲管浑浊肿胀减轻。结论:防己水煎液、多糖组分、生物碱组分能调节阿霉素诱导的肾病综合征模型大鼠低蛋白血症、血脂紊乱,降低蛋白尿含量,改善肾小球滤过功能,通过抑制炎症和氧化应激缓解肾脏损伤。多糖组分和生物碱组分治疗作用明显,为防己对阿霉素诱导的肾病综合征模型大鼠具有治疗作用的药效物质基础。
Objective:To observe the effects of Fangji(防己)water decoction and its various components on adriamycin-induced nephrotic syndrome in rats and explore the pharmacological basis of Fangji in the treatment of adriamycin-induced nephropathy.Methods:An adriamycin-induced nephropathy model was established in rats by tail intravenous injection of adriamycin.On the 1st and 8th days,rats were injected with adriamycin at doses of 4 mg/kg and 3.5 mg/kg,respectively,through the tail vein.Starting from the 11th day,rats were orally administered Fangji water decoction at 1.36 g/kg,polysaccharides at 0.82 g/kg,alkaloids at 0.19 g/kg,non-alkaloids at 0.17 g/kg,sterides at 0.03 g/kg,and positive control prednisone at 6.3 mg/kg for 14 consecutive days.The 24 h urinary protein,serum levels of ALB,TP,TC,TG,SCr,and BUN,levels of inflammatory factors IL-1β and TNF-α,oxidative stress markers MDA,GSH-Px,and SOD were detected,and histopathological changes in rat kidneys were measured by HE staining.Results:Compared with the normal control group,the model control group showed significantly increased urinary protein levels,elevated serum levels of SCr,BUN,TC,TG,and MDA,increased levels of IL-1βand TNF-α(P<0.01),decreased serum levels of ALB,TP,GSH-Px,and SOD,and increased renal index(P<0.01).Compared with the model control group,the Fangji water decoction at 1.36 g/kg,polysaccharides at 0.82 g/kg,and alkaloids at 0.19 g/kg showed varying degrees of increased serum ALB levels(P<0.05 or P<0.01),decreased serum TC and BUN levels(P<0.05 or P<0.01),reduced 24 h urinary protein concentration and serum TNF-αlevels(P<0.05 or P<0.01),decreased MDA levels(P<0.01),and increased GSH-Px levels and SOD activity(P<0.01).The polysaccharides at 0.82 g/kg also increased TP levels(P<0.01).The Fangji water decoction at 1.36 g/kg and polysaccharides at 0.82 g/kg reduced TG levels(P<0.05 or P<0.01).The Fangji water decoction at 1.36 g/kg and alkaloids at 0.19 g/kg reduced SCr levels(P<0.05 or P<0.01).The Fangji water decoction at 1.36 g/kg and polysacc
