摘要
目的通过不同比例浓缩牛乳蛋白及大豆分离蛋白饲料进行干预,研究不同来源蛋白对胎鼠及仔鼠小脑代谢组的影响。方法采用不同比例浓缩牛乳蛋白及大豆分离蛋白的饲料干预SD孕鼠,分为S1组(50%浓缩牛乳蛋白+50%大豆分离蛋白,n=24)、S2组(25%浓缩牛乳蛋白+75%大豆分离蛋白,n=24)、S3组(100%大豆分离蛋白,n=24)。干预从GD0.5持续至GD18.5或PD21。于GD19.5或PD21天,禁食后收集胎鼠小脑及仔鼠小脑,采用UPLC-MS行非靶向代谢组学检测。采用主成分分析观察不同组间样本的代谢轮廓差异;综合偏最小二乘判别分析和单因素统计方法,筛选组间差异代谢物,并进一步筛选不同组间共有的差异代谢物,对共有差异代谢物在不同组中的水平进行趋势方差分析。结果主成分分析表明S1与S3组胎鼠小脑代谢轮廓存在一定区分,S1与S2、S1与S3组仔鼠小脑代谢轮廓存在一定区分。综合偏最小二乘判别分析及单因素统计方法筛选差异代谢物后,S1与S2、S1与S3组仔鼠小脑间共发现11种共有的内源性差异代谢物,其中N-甲基组氨酸、4-氨基戊酸甜菜碱、1-甲基烟酰胺、鸟嘌呤、5-一磷酸胞苷、1-甲基腺苷等6种物质水平随大豆分离蛋白干预比例增加呈线性增加趋势。结论不同比例浓缩牛乳蛋白及大豆分离蛋白干预可对胎鼠及仔鼠小脑的代谢组产生影响。
Objective This study intends to explore the effects of different protein sources on the metabolome of the cerebellum of fetal and offspring rats by feeding different kinds of feed that contains different proportions of milk protein concentrate and soy protein isolate.Methods SD pregnant rats were intervened with feeds of different proportions of milk protein concentrate and soy protein isolate and divided into 3 groups,named S1(50%concentrated milk protein+50%soybean protein isolate,n=24),S2(25%concentrated milk protein+75%soybean protein isolate,n=24),and S3(100%soybean protein isolate,n=24).The intervention continued from gestational day 0.5 to gestational day 18.5 or postnatal day 21.On the gestational day 19.5 or postnatal day 21,the cerebellum of fetal and offspring rat was collected after fasting,and non-targeted metabolomics were detected by ultra-high performance liquid chromatography-mass spectrometry.Principal component analysis was used to observe the differences in metabolic profiles of samples between different groups;partial least squares discriminant analysis and single-factor statistical methods were combined to screen differential metabolites between groups,and further screen differential metabolites shared by different groups.Trend analysis of variance was performed on the levels of the substances in different groups.Results Principal component analysis showed that the metabolic profiles of the cerebellum of the fetal rat in S1 and S3 were distinct,and the metabolic profiles of the cerebellum of offspring rat in the S1 and S2,S1 and S3 were distinct.After comprehensive partial least squares discriminant analysis and single factor statistical method to screen differential metabolites,11 common endogenous differential metabolites were found in the cerebellum of offspring rat between different groups.The levels of six substances increased linearly in S1-S3,including N-methylhistidine,4-aminovaleric acid betaine,1-methylnicotinamide,guanine,cytidine 5′-monophosphate and 1-methyladenosine.Conclusio
