Chemical induced pluripotent stem cells and regenerative medicine

摘要 干细胞是具有增殖和分化潜能的细胞,可以分化为不同的组织和器官,是器官再生的重要基础。诱导多能干细胞技术可将分化终末端的细胞重编程为具有分化潜能的干细胞,诱导多能干细胞解决了胚胎干细胞使用的伦理限制,极大限度地扩展了干细胞在再生医学的应用。但是传统的导入转录因子(Oct4、Sox2、C-myc和Klf4)实现体细胞向多能干细胞的重编程技术由于复杂的操作和导入基因的突变风险限制了诱导多能干细胞的推广。近年来,不同的研究团队研发了操作简单、容易人为干预的化学小分子诱导重编程多能干细胞技术,进一步发展了诱导多能干细胞技术走向临床应用的可能。本文总结了化合物诱导重编程多能干细胞技术的进展,尤其是近来的小分子化合物诱导人多能干细胞的研究进展以及其在再生医学的应用现状及前景。 Stem cells are cells with proliferation and differentiation potential,which can differentiate into different tissues or organs.Stem cells are an important foundation for organ regeneration and future medical development.Induced pluripotent stem cell technology can reprogram the cells at the end of differentiation into stem cells with differentiation potential.Induced pluripotent stem cells solve the ethical restrictions on the use of embryonic stem cells,and greatly expand the application of stem cells in regenerative medicine.However,the traditional reprogramming technology of introducing transcription factors(Oct4,Sox2,C-myc and Klf4)to achieve somatic cell pluripotent stem cells has limited the promotion of induced pluripotent stem cells due to its complex operation and the mutation risk of imported genes.In recent years,different research teams have realized the small molecule induced reprogramming pluripotent stem cell technology,which is easy to operate and easy to be interfered by humans,and further developed the possibility of inducing pluripotent stem cell technology to clinical application.This review summarizes the progress of compound induced reprogramming of pluripotent stem cells,especially the recent research progress of small molecule compound induced human pluripotent stem cells,and its application status and prospects in regenerative medicine.
