摘要
以水杨苷含量及白柳皮提取物收率为评价指标,采用L9(34)正交试验设计,考察乙醇体积分数、料液质量体积比、提取时间、提取次数对白柳皮提取工艺的影响,筛选最佳提取工艺。利用HPLC法测定提取物中水杨苷的含量,以水杨苷含量及白柳皮提取物收率确定其最佳提取工艺为:4倍量的80%乙醇(体积分数)热回流提取2次,每次2 h。采用该工艺,白柳皮提取物中水杨苷含量为16.5%(质量分数),高于市售产品。综上,优化后的提取工艺稳定可靠,适合工业化生产,可为白柳皮资源开发利用提供参考。
The content of the salicin and the white willow bark extract yield were chosen as the evaluation indicators,and the L9(34)orthogonal test was used to investigate the effects of ethanol volume fraction,ratio of solid-liquid,extraction time,extraction times on the extraction process of white willow bark,and then choose the optimum extraction process.HPLC method was used to determine the content of salicin in the extract.Based on the content of the salicin and the white willow bark extract yield,the optimum extraction process was determined as follows:the solid-liquid ratio at 1:4,ethanol vloume concentration is 80%and reflux extract twice,each for 2 h.Using this process,the mass content of the salicin in white willow bark extraction was 16.5%,higher than that of commercially available products.In summary,the optimum extraction process is stable,feasible,and suitable for industrial production.It can provide reference for the development and utilization of white willow bark resources.
