摘要
目的:系统研究鼠曲草的化学成分,并探讨其发挥治疗慢性阻塞性肺疾病(COPD)的主要药效成分和潜在作用机制。方法:运用UPLC-QE-MS技术,通过XCMS软件对鼠曲草提取物正、负离子质谱数据进行分析,应用TCMSP数据库获取鼠曲草活性成分及靶点;以GeneCards和OMIM数据库检索、获取疾病靶点,使用Venny 2.1.0找出药物-疾病共同靶点。应用Metascape数据库对共同靶点进行GO功能富集分析、KEGG通路富集分析。结果:在Composite Score>0.99的情况下UPLC-QE-MS共鉴定出化合物133个,其中正离子模式鉴定82个成分、负离子模式鉴定51个。基于各数据库收集,共获得10个鼠曲草活性成分及185个可用于后续分析的靶点,1637个COPD作用靶点(Relevance score≥20),交集靶点共91个。GO生物过程共449条,包括BP有23条、MF 128条、CC 91条;KEGG相关信号通路包括p53信号通路、PI3K-Akt信号通路、IL-17信号通路、HIF-1信号通路、MAPK信号通路等。结论:采用UPLC-QE-MS技术进行定性分析,方法稳定可靠,为鼠曲草的质量控制提供了一定的参考依据。网络药理学研究表明,鼠曲草防治COPD的作用机制具有“多成分、多通路、多靶点”的特点,研究结果可为阐明鼠曲草的药理作用机制提供重要参考。
Objective:To systematically study the chemical components of Gnaphalium affine D.Don and their possible effective components and potential mechanism in the prevention and treatment of chronic obstructive pulmonary disease(COPD).Methods:UPLC-QE-MS technology and XCMS software were used to analyze the positive and negative ion mass spectrometry data of The extract of Gnaphalium affine D.Don,and TCMSP database was used to obtain the active components and targets of Gnaphalium affine D.Don.GeneCards and OMIM databases were used to retrieve and obtain disease targets,and Venny 2.1.0was used to find drug-disease common targets.GO functional enrichment analysis and KEGG pathway enrichment analysis were performed on the common targets using Metascape database.Results:Under the condition of Composite Score>0.99,a total of 133compounds were identified by UPLC-QE-MS,of which 82were identified by positive ion mode and 51by negative ion mode.A total of 10active components,185targets,1637COPD targets(Relevance score≥20)and 91intersection targets were obtained from various databases.There were 449GO biological processes,including 230BP,128MF and 91CC.KEGG related signaling pathways include p53signaling pathway,PI3K-Akt signaling pathway,IL-17signaling pathway,HIF-1signaling pathway,MAPK signaling pathway and other signaling pathways.Conclusion:UPLC-QE-MS technique was used for qualitative analysis.The method was stable and reliable,which provided a certain reference for the quality control of Gnaphalium affine D.Don Network pharmacology studies showed that the mechanism of action of Gnaphalium affine D.Don in the prevention and treatment of COPD was characterized by“multiple components,multiple pathways and multiple targets”.The results provide important refernce for further elucidating the pharmacological mechanism of Gnaphalium affine D.Don.
