摘要
探究黑莓果渣花青素的体内降血糖、降血脂和抗氧化作用并初步阐明其作用机制。超声波辅助提取黑莓果渣花青素后经聚酰胺树脂纯化得到精制黑莓果渣花青素,采用2型糖尿病小鼠模型研究其降血糖、降血脂及抗氧化能力。结果表明黑莓果渣花青素具有一定的降血糖、降血脂及抗氧化能力。在28天的服用周期内,黑莓果渣花青素能显著降低小鼠的空腹血糖还可以防止因糖尿病导致的体重下降,其机制是通过调节INS分泌及肝糖原的合成以达到降血糖的目的。黑莓果渣花青素可以显著降低TG、TC和LDL-C含量,提高HDL-C含量以调节血脂来达到降血脂的目的,还可以通过提高具有清除和修复氧化自由基的抗氧化酶类(SOD、GSH-px和CAT)活性和抑制脂肪氧化酶来阻碍MDA生成来提高抗氧化活性。
The purpose of this paper is to explore the hypoglycemic,hypolipidemic and antioxidant effects of anthocyanins from blackberry pomace in vivo and preliminarily clarify its mechanism.Ultrasonic assisted extraction of anthocyanins from blackberry pomace was purified with polyamide resin to obtain refined anthocyanins.The hypoglycemic,hypolipidemic and antioxidant abilities of anthocyanins were studied in type 2 diabetes mice.The results showed that anthocyanins had certain hypoglycemic,hypolipidemic and antioxidant abilities.During the 28 days administration period,anthocyanins could significantly reduce the fasting blood-glucose of mice and prevent mice weight loss caused by diabetes.The mechanism was to reduce blood-glucose by regulating the secretion of INS and the synthesis of liver glycogen.Anthocyanins could significantly reduce the contents of TG,TC and LDL-C,increase HDL-C to regulate blood lipids to achieve the purpose of reducing blood lipids.The antioxidant activity was improved by increasing the activities of antioxidant enzymes(SOD,GSH-px and CAT)that scavenged and repaired oxidative free radicals,inhibiting the production of MDA by inhibiting the activity of fat oxidase.
