摘要 目的提取浙贝母花中的不同化学成分,分析其体内抗氧化能力。方法采用超声提取法和萃取法提取浙贝母花中的不同化学成分。皮下注射D-半乳糖诱导小鼠氧化衰老模型。随机将小鼠分为7组:空白组、D-半乳糖诱导模型组、维生素C阳性对照组、浙贝母花给药组(乙醇提取物组、石油醚萃取物组、乙酸乙酯萃取物组、正丁醇萃取物组)。连续皮下注射、灌胃4周后,眼球取血并处死,解剖取其肝脏、脑组织、脾脏和胸腺组织,测定肝脏、脑组织抗氧化指标。结果与对照组相比,模型组小鼠脏器指数均降低(P<0.05或P<0.01),血清、肝脏和脑组织中SOD水平降低(P<0.01),肝脏和脑组织中CAT水平降低(P<0.01或P<0.001),血清、肝脏和脑组织中MDA水平提高(P<0.01),肝脏和脑组织GSH-PX水平降低(P<0.01或P<0.001)。与模型组比较,给药组小鼠脏器指数、SOD水平、CAT水平、GSH-PX水平均提高,MDA水平均降低(P<0.05)。结论浙贝母花浸提物中不同物质成分在增加实验小鼠抗氧化酶活性、保护机体内的还原性物质不被氧化、减少脂质过氧化产物、延缓氧化过程等方面均有显著作用。 Objective To extract different chemical constituents from Fritillaria thunbergii Miq and to study the antioxidant capacity of different chemical constituents in Fritillaria thunbergii Miq.Methods Ultrasonic extraction and extraction methods were used to extract different chemical constituents from Fritillaria thunbergii Miq.Subcutaneous injection of D-galactose was used to induce oxidative aging model in mice.The mice were randomly divided into 7 groups:the control group,the D-galactose-induced model group,the vitamin C positive control group,and the Fritillaria thunbergii Miq administration group(Fritillaria thunbergii Miq ethanol extract group,Fritillaria thunbergii Miq ethyl acetate extract group,Fritillaria thunbergii Miq petroleum ether extract group,and Fritillaria thunbergii Miq n-butanol extract group).After 28 days of continuous subcutaneous injection and gavage,blood was taken from the eyes and executed.The liver,brain tissue,spleen and thymus tissues were taken by autopsy,and the antioxidant indexes of liver and brain tissues were measured to study the antioxidant capacity of different material components of Fritillaria thunbergii Miq extracts.Results Compared with the control group,the organ indices of mice in the model group were all reduced,with statistical significance(P<0.05 or P<0.01),SOD levels in serum,liver and brain tissues were significantly reduced(P<0.01),CAT levels in liver and brain tissues were significantly reduced(P<0.01 or P<0.001),MDA levels in serum,liver and brain tissues were significantly increased(P<0.01),liver and GSH-Px levels in brain tissue were significantly decreased(P<0.01 or P<0.001).Compared with the model group,the organ indices of mice in the administered group were all increased,the SOD levels were all increased,the CAT levels w-ere all increased,the MDA levels were all decreased,and the GSH-Px levels were all increased(P<0.05).Conclusion The different substance components in the extract of Fritillaria thunbergii Miq has significant effects on the antioxidant effect
