摘要
目的:建立广西产不同品种石韦药材的HPLC指纹图谱,对其进行分析和质量评价,同时测定石韦中绿原酸、芒果苷、山奈素、芦丁的含量。方法:采用HPLC测定广西产6批不同品种石韦药材的含量,分别运用SPSS 19.0软件和中药色谱指纹图谱相似度评价系统2004 A版对广西产6批不同品种石韦药材的14个共有峰进行聚类分析(HCA)、主成分分析(PCA)和相似度分析。结果:广西产不同品种石韦中绿原酸含量最高,其次为芦丁、芒果苷和山奈素。其中,有柄石韦中绿原酸最高,石韦中芒果苷最高,而在柔软石韦中芦丁及山奈素含量较高。对其指纹图谱进行比较,聚类分析中除绒毛石韦外,其它石韦可为一类;主成分分析中越石韦、庐山石韦、有柄石韦和柔软石韦为一类;在相似度分析中,对比药典项下石韦,可见中越石韦与庐山石韦、有柄石韦较接近。结论:广西产不同品种石韦中绿原酸,芒果苷,山奈素,芦丁含量存在差异。此外,相似度分析,主成分分析和聚类分析结果为鉴别石韦药材提供依据,为石韦药材质量标准的完善和临床用药提供参考。
Objective The HPLC Fingerprints were built to analyze and evaluate the different Pyrrosia from Guangxi province.Simultaneously,the contents of chlorogenic acid,mangiferin,kaempferol and rutin were determined.Methods 14 peaks of the common model were collected and used by establishing HPLC fingerprint from Guangxi Pyrrosia species The data were applied to professional software named SPSS 19.0 statistical analysis software for hierarchical cluster analysis(HCA)and principal component analysis(PCA)and Similarity Evaluation System for chromatographic fingerprint of TCMs(Version 2004 A)recommended by the SFDA of China for evaluating similarities.Results The content of four ingredients were determined,which it were chlorogenic acid>rutin>mangiferin>kaempferol.Among them,chlorogenic acid and mangiferin were the highest in P.petiolosa and P.lingua respectively,while the contents of rutin and kaempferol were higher in P.mollis.To compare their fingerprints,except for P.subfurfuracea,the others can be classified as one class by HCA.The PCA method showed that it was categorized into one group:P.tonkinensis,P.sheareri,P.petiolosa and P.mollis.And the similarity values,P.tonkinensis,P.sheareri,and P.petiolosa,were very close by comparing the pharmacopeia.Conclusions The content of main ingredients(chlorogenic acid,mangiferin,kaempferol and rutin)was difference from different species Pyrrosia.Besides,it provides a basis for identifying and evaluating the quality of Pyrrosia by the similarity analysis,PCA and HCA,and offers a reference for the quality standards and clinical application of Pyrrosia.
