摘要
目的 旨在分析利胆清胰汤通过调节Nf-κB信号通路对小鼠急性胰腺炎模型的影响。方法 2020年10月—2021年4月在牡丹江医学院临床学院将30只C57BL/6雄性小鼠随机分为正常组、模型组和利胆清胰汤组,每组10只,通过8次腹膜内注射雨蛙素构建小鼠急性胰腺炎模型,给予利胆清胰汤干预处理后,对小鼠疼痛行为进行观察,检测血清淀粉酶和髓过氧化物酶(myeloperoxidase, MPO)水平,同时观察胰腺组织病理改变,免疫印迹法检查胰腺组织中核因子κB(nuclear factor kappa-B, NF-κB)、NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor thermal protein domain associated protein 3, NLRP3),半胱氨酸蛋白酶-1(caspase-1),凋亡相关斑点样蛋白(apoptosis-associated spot-like proteins, ASC),磷酸化NF-κB-p65和非磷酸化p65蛋白表达水平。结果 与对照组小鼠相比,模型组小鼠疼痛行为第一次出现的时间明显缩短,血清淀粉酶、MPO水平明显升高,胰腺组织损伤明显升高,NLRP3、ASC、Caspase-1、p-NF-κB p65表达水平明显升高,差异有统计学意义(P<0.05)。与模型组相比,利胆清胰汤组小鼠疼痛行为第一次出现明显延长,胰腺组织损伤明显减轻,血清淀粉酶、MPO水平明显降低,差异有统计学意义(P<0.05)。模型组NLRP3、ASC、Caspase-1、p-NF-κBp65表达水平分别为(1.47±0.15)、(1.19±0.10)、(1.89±0.26)、(0.57±0.10)与利胆清胰汤组的(0.99±0.12)、(0.77±0.12)、(0.92±0.10)、(0.24±0.08)比较,差异有统计学意义(P<0.05)。但各组小鼠非磷酸化NF-kB-p65蛋白表达水平比较,差异无统计学意义(P>0.05)。结论 利胆清胰汤可能通过调节抑制NF-κB通路与NLRP3炎性小体激活对AP产生预防和治疗作用。
Objective To analyze the effect of Lidan Qingyi Decoction on acute pancreatitis model in mice by regulat‐ing Nf-kB signaling pathway.Methods From October 2020 to April 2021,30 male C57BL/6 mice were randomly di‐vided into normal group,model group and Lidanqingyi Decoction group,with 10 mice in each group.A mouse model of acute pancreatitis was constructed by 8 intra peritoneal injections of hyranin.After the intervention of Lidanqingyi Decoction,the pain behavior of the mice was observed.The levels of serum amylase and myeloperoxidase(MPO)were detected,and the pathological changes of pancreatic tissue were observed.Detection of nuclear factor kappa-B in pan‐creatic tissue by Western blotting NF-κB,NOD-like receptor thermal protein domain associated protein 3(NLRP3),caspase-1,NOd-like receptor thermal protein domain associated protein 3,Expression levels of apoptosis-associated spot-like proteins(ASC),phosphorylated NF-B-p65 and non-phosphorylated p65 proteins.Results Compared with the control group,the time for the first occurrence of pain behavior was significantly shortened,the levels of serum amylase and MPO were significantly increased,the pancreatic tissue damage was significantly increased,and the ex‐pression levels of NLRP3,ASC,Caspase-1 and P-NF-B p65 were significantly increased in the model group,the dif‐ference was statistically significant(P<0.05).Compared with the model group,the pain behavior of mice group was sig‐nificantly prolonged for the first time,the damage of pancreatic tissue was significantly alleviated,and the levels of se‐rum amylase and MPO were significantly decreased in the Lidanqingyitang,the difference was statistically significant(P<0.05).The expression levels of NLRP3,ASC,Caspase-1,and p-NF-B-p65 in the model group were(1.47±0.15),(1.19±0.10),(1.89±0.26),and(0.57±0.10),respectively,compared with those in the Lidan Qingyi Tang group(0.99±0.12),(0.77±0.12),(0.92±0.10),and(0.24±0.08),the difference was statistically significant(P<0.05).There was no sta‐tistic
