摘要
为了更好地利用蛹虫草资源,对鲜蛹虫草、干蛹虫草水提物的细胞毒性和抗氧化能力进行评价。按照体质量分别对6组试验小鼠腹腔注射200 mg/kg鲜蛹虫草水提物、200 mg/kg干蛹虫草水提物、10 mL/kg无菌水、4 mg/kg丝裂霉素C、200 mg/kg鲜蛹虫草水提物+4 mg/kg丝裂霉素C和200 mg/kg干蛹虫草水提物+4 mg/kg丝裂霉素C,测定试验组和对照组小鼠骨髓中嗜多染红细胞与成熟红细胞的比值,用于评价鲜、干蛹虫草的细胞毒性。按照体质量对阴性对照组小鼠灌饲生理盐水,模型对照组小鼠先后腹腔注射80 mg/kg的环磷酰胺并灌饲生理盐水,对试验组小鼠分别先后腹腔注射80 mg/kg的环磷酰胺及灌饲100 mg/kg、200 mg/kg的鲜蛹虫草和干蛹虫草水提物,测定对照组和试验组小鼠心脏、肝脏和肾脏组织中超氧化物酶歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶和丙二醛的含量,用于评价鲜、干蛹虫草抗氧化能力。试验结果表明,鲜、干蛹虫草水提物对细胞无遗传毒性,并具有抗细胞毒性和抗氧化能力,而且鲜蛹虫草抗氧化能力强于干蛹虫草,更适宜作为功能性食品资源。
In order to make better use of Cordyceps militaris resources,the cytotoxicity and antioxidant capacity of fresh Cordyceps militaris and dry Cordyceps militaris aqueous extracts were evaluated.Six groups of mice were intraperitoneally injected with 200 mg/kg fresh Cordyceps militaris aqueous extract,200 mg/kg dry Cordyceps militaris aqueous extract,10 mL/kg sterile water,4 mg/kg mitomycin C,200 mg/kg fresh Cordyceps militaris aqueous extract+4 mg/kg mitomycin C and 200 mg/kg dry Cordyceps militaris aqueous extract+4 mg/kg mitomycin C,respectively.The ratio of polychromatic erythrocytes to mature erythrocytes in the bone marrow of mice in the experimental groups were determined to evaluate the cytotoxicity of fresh and dry Cordyceps militaris.According to the body mass,the mice in the negative control group were fed with normal saline,and the mice in the model control group were intraperitoneally injected with 80 mg/kg cyclophosphamide and then fed with normal saline.The mice in the experimental groups were intraperitoneally injected with 80 mg/kg cyclophosphamide and then fed with 100 mg/kg and 200 mg/kg fresh and dry Cordyceps militaris aqueous extract.The contents of superoxide dismutase,catalase,glutathione peroxidase and malondialdehyde in the heart,liver and kidney tissues of the mice in the control group and the experimental groups were measured to evaluate the antioxidant capacity of fresh and dry Cordyceps militaris.The results showed that the aqueous extracts of fresh and dry Cordyceps militaris had no genotoxicity to cells,and had anti-cytotoxicity and antioxidant capacity.The antioxidant capacity of fresh Cordyceps militaris is stronger than that of dry Cordyceps militaris,which is more suitable as a functional food resource.
