通过转录组测序挖掘三叶青黄酮类物质生物合成的相关基因

摘要 目的:通过转录组测序挖掘三叶青黄酮成分生物合成相关基因。方法:三叶青叶经过Illumina HiSeq 4000高通量测序,获得转录组数据,通过数据组装、拼接及注释,挖掘三叶青黄酮类物质生物合成的相关基因。结果:5个三叶青样品获得111.281Gb数据,使用trinity组装,得到626199个transcripts,获得396496个unigenes,将unigenes与NR、Swiss-prot、STRING、GO、KEGG、Pfam这6个数据库进行对比分析。分析KEGG代谢通路,获得3条与黄酮物质合成的代谢通路。黄酮类物质生物合成的相关基因有:黄酮类31条,异黄酮类2条;黄酮和黄酮醇3条。与三叶青黄酮成分合成相关的酶有:查尔酮合成酶(CHS),查尔酮异构酶(CHI),柚皮素-3-双加氧酶(F3H)等九种酶。结论:首次基于三叶青转录组测序数据,获得三叶青黄酮类成分生物合成的关键酶基因,为后续三叶青基因功能的研究提供数据支持。 Objective To identify flavonoid biosynthesis-related genes in Trifolium pratense using transcriptome sequencing.Methods Transcriptome data of Trifolium pratense were obtained through Illumina HiSeq 4000 high-throughput sequencing.The data were assembled,spliced,and annotated to identify genes related to the flavonoid metabolic pathway.Results A total of 111.281 Gb data were obtained from five Trifolium pratense samples after sequencing.Trinity assembly yielded 626,199 transcripts and 396,496 unigenes.The unigenes were compared with six databases:NR,Swiss-prot,STRING,GO,KEGG,and Pfam.KEGG metabolic pathway analysis revealed three metabolic pathways related to flavonoid biosynthesis in Trifolium pratense:flavonoid biosynthesis with 31 related genes;Isoflavonoid biosynthesis with two related genes;And flavone and flavonol biosynthesis with three related genes.Enzymes related to flavonoid synthesis in Trifolium pratense include chalcone synthase(CHS),chalcone isomerase(CHI),flavanone 3-hydroxylase(F3H),and six other enzymes.Conclusion For the first time,key enzyme genes for flavonoid biosynthesis in Trifolium pratense were obtained based on transcriptome data,thereby providing data support for subsequent studies of gene function in Trifolium pratense.
